Vitamin C (ascorbic acid) is a well-known, water-soluble nutrient that readily acts as a strong reducing agent. It is an essential component of most living tissues. Apart from antioxidant activity, it has been reported to inhibit cancer cell growth in vitro in human cancer cells. Although, the effect of vitamin C in cancer treatment remains controversial, a high intake of fresh fruits and vegetables which contain vitamin C is associated with a reduced risk for most types of cancer. Therefore, the present study was design to study the anticancer and anti-proliferative activity of Vitamin C (ascorbic acid) on gastric cancer cell line AGS. A Gulo-/- mice model like human (it can’t synthesis vitamin C) was also investigated for protection effect of vitamin C and biomarkers of gastric cancer by a proteomic approach. Vitamin C showed a cytotoxic effect on human gastric cancer cell line AGS (LD50 300 µg/ml). Flow cytometry analysis showed that vitamin C increased the sub-G1 (apoptosis) population and apoptosis confirmed by fluorescein isothiocyanate-Annexin V double staining in AGS cells. Moreover, specific immuno-blotting revealed the expression of the phosphorylated form of Bad (S136), 14-3-3σ, pro-caspases-3, -6, -8, and-9 protein levels were significantly decreased and Bax/Bcl-xL ratio was increased in a dose-dependent manner. Wound healing assay results showed that vitamin C inhibited AGS cell proliferation. These findings revealed that vitamin C at pharmacological concentrations (300µg/ml) induces apoptosis in AGS cells mainly through the down-regulation of 14–3-3σ protein and dephosphorylates Bad proteins, which increase the Bax/Bcl-xL ratio, resulting in apoptosis via a mitochondrial dependent pathway. Further, proteome analysis performed on the AGS cells treated with vitamin C at pharmacological concentrations (300µg/ml). Differentially expressed proteins were identified by matrix-assisted laser desorption/ionization time-of-flight and mass spectrometry (MALDI-TOF/MS). Proteomic analysis revealed 20 differentially expressed proteins, of which six were up-regulated and fourteen were down-regulated. In particular, cytoskeletal and signal transduction related proteins were down-regulated, and detoxification and redox proteins were up-regulated in the vitamin C-treated AGS cells compared with the control. The expressions of 14-3-3 isoforms were also verified with immuno-blotting analysis. These proteome results revealed that vitamin C inhibited the cell growth, and apoptosis related proteins were involved in promoting and regulating cell death in AGS cells. Taken together, in vitro research findings suggest that vitamin C might be a potential therapeutic agent for gastric cancer. Unlike most other mammals and animals, human bodies do not have the ability to synthesis vitamin C inside of their own bodies. Therefore, humans must obtain vitamin C through daily diet. With recent advances in proteomics protein biomarker discovery is become one of the of proteome research. Screening methods of proteome by MALDI-TOF/MS analyses of animal samples like stomach tissues, liver and serum are promising technologies for biomarker discovery. A Gulo-/- mice model was also investigated for the discovery of biomarkers of gastric cancer by a proteomic approach. The knock-out mice were divided into three groups; control group, Helicobacter pylori-infected group and the group of H.pylori infection followed by Diethylnitrosamine (DENA) treatment. All these three groups were supplemented vitamin C (20mg/animal/day, PO) and only group 3 treated DENA (10mg/L, PO). After 48 weeks, all animals were sacrificed and serum, stomach and liver were collected. Immunohistochemistry of stomach tissue showed that a tumor suppressor p53 was decreased, and 14-3-3ε and 14-3-3δ expressions were increased in H. Pylori-infected group and H. Pylori +DENA group when compare with the control group. However, little difference was found in p53 and no difference in 14-3-3ε and 14-3-3δ expression in the liver tissue.
