The purpose of this research were production of bio-energy and alginate-degrading enzyme from Laminaria japonica. Therefore, production of bio-ethanol from the main sugar component of Laminaria japonica. A marine alginate-degrading bacterium was isolated and characterized. Alginate-degrading bacteria and sludge were co-fermented to produce bio-gas from the by-product of bio-ethanol product. Firstly, Ethanol producing microorganism were obtained from the Korea Culture Center of Microorganisms (http://www.kccm.or.kr/) and Korean Collection for Type Culture (http://kctc.kribb.re.kr/). Zymomonas mobilis KCTC 1534 and Paracoccus sp. used the main sugar components of Laminaria japonica. Ethanol production by Zymomonas mobilis KCTC1534 reached 2.313 g/L, 1.316 g/L, 2.336 g/L and 2.196 g/L from 10 g/L of alginate, laminaran, mannitol and from 10 g/L of Laminaria japonica medium, respectively. Paracoccus caeni produced the highest bio-ethanol among all tested bacterial strains, reaching 3.240 g/L from 10 g/L of mannitol and 4.153 g/L from 40 g/L of mannitol. The main sugar components of Laminaria japonica were not used by several microorganism. But, ethanol production detected from mannitol. Secondly, alginate-degrading strains were isolated from Namhae Naeng-Cheon mud flat and Tongyeong sea. alginate-degraing strain was named Sanguibacter keddieii NC9 after 16S rDNA sequencing. The optimum culture condition for the degradation of alginate-degrading was sodium alginate 10 g/L, peptone 5 g/L, 40℃, pH 9 and 36 hours incubation time. Thirdly, biogas was produced from the by-product of ethanol fermentation of Laminaria japonica through anaerobic fermentation using Sanguibacter keddieii NC9, Erwinia tasmaniensis and Sludge. The optimal ratio of sludge and Sanguibacter keddieii NC9 was 1:2 for bio-gas production, and the hydrogen and methane gas production levels under these conditions were 1575.86 mL/L and 4339.14 mL/L, respectively.
