Mesophilic batch reactors were used to investigate the biochemical methane production potentials (BMP) of piggery and agricultural wastes, and mesophilic semi-continuous stirred tank reactors (CSTR) were applied to measure the methane production yield from piggery waste. Moreover, automatic facility for the continuous measurement of gas production was invented and attached to the CSTR. To perform the BMP tests of piggery and agricultural wastes, three kinds of substrates were added to serum vials (500 ml) with sludges from preliminary experiment as an inoculum. Reactor A was prepared with 60 ml of sludge and 240 ml of piggery waste. Reactors B and C were prepared with the same contents of reactor A, but added with rice straw (15 g) and peanut hull (15 g), respectively. At reactor A, biogas was not produced after 30 days of incubation and the total amount of gas production was only 0.9 L. However, reactor B with piggery waste and rice straw produced much amounts of biogas; the cumulative amounts were 4.6 L at 30 days and 5.9 L at 55 days of incubation and the biogas was no longer produced at the incubation time of 55 days. Thus, the contribution of rice straw to the biogas production were supposed to be 3.8 L during the first 30 days and 5.0 L during the total incubating period. At reactor C with piggery waste and peanut hulls, biogas was produced until 41 days of incubation and the cumulative amount was 3.2 L and the contribution of peanut hull for the biogas production was supposed to be 2.3 L. The maximum methane contents of biogas from reactors A, B, and C were 55.8%, 67.0%, and 45.6%, respectively. Substrate removal rates which were converted to methane from reactors A, B, and C were 1.26, 7.46, and 4.49 L-CH4/g-VS, respectively, and the methane yields were 0.09, 0.12, and 0.08 L-CH4/g-VS, respectively. Two sets of CSTR were prepared with 8 L of piggery waste and 2 L of sludge were added and stand the first 27 days of operation to establish the stabilization and adaption of inoculum to the fresh substrate. After that time, 500 ml of working volume was discarded and the same amount of fresh piggery waste was added to the reactors. The average volume of gas counter manufactured for this studies was 55 ml/count. The differences of maximum amount of biogas production per day were somewhat large, 2.7 and 6.7 L/d. However, the average methane contents of two sets of CSTR were 57.7 and 68.6%, and the maximum contents were 86.1 and 87.0%.
