Lactococcus lactis BFE920, a nisin-producing lactic acid bacteria (LAB), is a well-known probiotic that can stimulate the mucosal immunity of human and mouse models. Also, the utilization of the LAB as a vaccine delivery vehicle, using its nisin-controlled expression (NICE) system, has been explored in the mouse system, but there have been no published works on its use in the fish system. This led us to propose a possible LAB vaccine system using L. lactis BFE920 that works against Streptococcus iniae infection in fish, one of the most problematic fish pathogens in aquaculture of flounders. We cloned the simA gene (M-like membrane protein) and the sip11 gene (putative iron-binding protein) from S. iniae into three different vectors that express this protein in intracellular (pLB23), secreted (pLB11) and cell wall-anchored (pLB113) form in L. lactis BFE920 together with the HA tag attached to the end of the sequence. The cloning and production of the plasmid construct was successful as confirmed by SimA and Sip11 specific primer PCR and plasmid sequencing. S. iniae specific polyclonal antibodies were generated in mouse and used to detect simA proteins from recombinant L. lactis BFE920 by western blotting.
