Before the SNP analysis, 48 LRs (26 males and 22 females) aged in 1-15 years old were assigned into two groups based on British veterinary association/kennel club (BVA/KC) scores by X-ray measurements: normal dogs (0-10) and CHD dogs (≥11). Optimal SNP markers for CHD were found from 11 sites of chromosome 4, 4 sites of chromosome 38, and 10 sites of chromosome X using a 22K canine SNP chip (genome-wide P < 0.001). These SNP sites were detected in all aged LRs judged to CHD by X-ray diagnosis, which was unrelated to sexual differences. Furthermore, in order to search fast and more useful CHD markers in clinic fields, the restriction fragment length polymorphism (RFLP) experiments were carried out 48 LRs using 11 of 25 SNPs detectable by restriction enzymes. Consequently, only a SNP (BICF2S23030416) was found as a useful marker to distinguish CHD in LRs. These findings indicate that SNP and RFLP methods may be a useful technique for CHD diagnosis of LRs before puberty.
