In vitro 에서 AF와 AFG의 자당의 분해효소인 Sucrase의 저해효과는 각각 높은 저해효과를나타났다. 두 아마도리 전위 화합물의 Sucrase IC50은 각각 AF 6.4 mM AFG 6.2 mM 인 것으로 나타났다. α-glucosidase Inhibitor 의 대표적인 약물인 Acarbose® 부작용의 지표로 사용되는 α-amylase의 활성을 측정한 결과, AF와 AFG 두 물질 모두 α-amylase의 저해효과가 낮은 것으로 나타났다. 두물질의 α-amylase IC50은 AF는 36.3 mM AFG의 경우 37.6 mM로 나타났다. Sprague-Dawley (SD) rat을 이용한 in vivo 식후 혈당 상승억제 실험을 통하여 AF와 AFG (0.10, 0.50 g/kg body weight)의 농도에서 starch, sucrose에 대해서 혈당상승 억제 효과는 대조군에 비해 혈당상승 억제효과가 유의적으로 경감된 것을 확인 할 수 있었다. 2형 당뇨병이 유발된 db/db mouse을 이용한 6주간의 장기 feeding test를 실시한 결과 Acarbose® 투여군 (0.4 g/kg diet weight)과 AF 투여군 (40 g/kg diet weight)에서 db/db mouse의 체중이 control 군이경우 42.8 ± 2.7단위 체중이 증가 하였으나, AF 투여군과 Acarbose® 투여군의 경우 37.4 ± 1.5 g 와 38.0 ± 4.2 g 비슷한 수준으로 체중경감 효과가 나타났다. 6주후 혈중 포도당 농도 또한 Acarbose® 투여군과 AF투여군의경우 223.8 ± 77.0 mg/dL 와 264.2 ± 21.5 mg/dL 로 대조군 558.6 ± 57. 보다 현저히 낮은 수준을 나타냈다. 또한 장기간의 혈중 포도당 농도를 나타내는 Hemoglobin A1c (HbA1c) 경우 또한 Acarbose® 투여군과 AF 투여군은 5.1 ± 0.3 %, 7.1 ± 0.6 %로 대조군의 결과인 9.8 ± 1.0 %보다 낮은 수치를 나타내어 6주간 대조군보다 낮은 혈중 포도당 농도를 유지했음을 알수 있다. 그리고 Acarbose® 투여군의 경우 Acarbose®의 대표적인 부작용인 맹장의 무게가 1.60 ± 0.40 g 으로 대조군과 AF 투여군의 0.28 ± 0.09 g, 0.27 ± 0.07 g 보다 증가한것으로 나타났으며, AF투여군의 경우는 대조군과 비슷한 부피와 무게를 나타냈다. 따라서 AF는 Acarbose®와 같은 혈당상승 억제효과가 있으며, Acarbose® 에서 나타나는 부작용 또한 적은것으로 사료된다. 급성 독성을 조사하기 위해서 SPF 렛트 수컷 5마리 암컷 5마리에 AF를 경구를 통하여 단회 투여하고 15일동안 증상관찰, 사망동물관찰, 사망동물관찰, 체중변화와 측정및 부검 소견을 관찰하였다. 투여 용량은 G1 (0 mg/kg), G2(2 500 mg/kg), G3(5 000 mg/kg), G4(10 000 mg/kg) 으로 하였다. 그결과 투여후 고농도에서 착색변이 확인되었고 15일동안 사망동물이 없었으며 체중의 변화가 대조군과 비교하여 차이가 없었으며, 부검결과 또한 별다른 이상변화가 없었다. 따라서 AF의 경우 고농도 급성독성이 없는것으로 나타났다. 아급성 독성을 조사하기 위해서 SPF 렛트 수컷 10마리 암컷 10마리에 AF를 경구를 통하여 3달 동안 투여하고 증상, 사망동물, 체중변화와 측정및 부검 소견을 관찰하였다. 투여 용량은 0, 500, 800, 2 000 mg/kg으로 하였다. 그결과 단회투여 시험결과와 같이 투여후 고농도에서 착색변이 확인되었고 3달 동안 사망동물이 없었으며 체중의 변화가 대조군과 비교하여 차이가 없었으며, 부검결과 또한 별다른 이상변화가 없었다. 따라서 AF의 경우 아급성독성이 없는것으로 나타났다. 이상의 연구결과로 AF는 α-glucosidase저해활성을 통해 식후혈당상승을 억제하는 소재로서 부작용과 독성이 없는 안전한 물질이며 향후 인체임상시험을 통해 효능을 검증하여 혈당상승억제 건강기능식품소재로서의 개발이 가능할 것으로 사료된다.

This is the first report of the mode of action and effect of AF and AFG on the type 2 diabetes management related inhibition of postprandial hyperglycemia in vitro and in animal model. In our previous study, standards AF and AFG were chemically synthesized and in this study their inhibitory activities against rat intestinal α-glucosidases and porcine pancreatic α-amylase were investigated in vitro. The IC50 value of the in vitro inhibitory activity of AF and AFG on rat intestinal sucrase was high and in similar levels (6.40 and 6.20 mM, respectively). Additionally, a mild pancreatic α-amylase inhibitory activity was observed, with IC50 values 36.30 and 37.60 mM for AF and AFG, respectively. The effect of AF and AFG on the postprandial blood glucose increase after meal was investigated in Sprague-Dawley rats fed on starch or sucrose meals. Both amadori compounds significantly reduced the postprandial blood glucose levels after starch or sucrose loading. These results indicate that AF and AFG, maillard reaction products, may have anti-diabetic effect by suppressing carbohydrate absorption in the gastrointestinal level, and thereby reducing the postprandial increase of blood glucose. To test whether arginyl-fructose (AF) play a role in the blood glucose lowering in db/db mice model, db/db mice were randomly assigned to high-carbohydrate diets (66.1% corn starch), in AF contents (4% in diet) for 6 weeks. Changes in body weight, blood glucose level, and food intake were measured daily for 42 days. Administration of AF resulted in a significant decrease of blood glucose level and body weight. The level of HbA1c relevant to seriousness of diabetes was also significantly decreased during 6 weeks. Administration of Acarbose® (0.04% in diet), positive control, also significantly alleviated the level of blood glucose, HbA1c, and body weight. These results indicate that AF maillard reaction product may have anti-diabetic effect by suppressing carbohydrate absorption in the gastrointestinal level. The present study was carried out to assess the toxicity of AF in Sprague-Dawley Rats following a single oral administration. Compound-colored stool and diarrhea were presumed to be caused by physical stimuli due to dosing of excessive amount of test article in a fasting state, because there was no significant change in body weight. The high dose level was chosen to provide significant exposure to the test material and the other dose levels were set to provide data adequate for risk assessment purposes. There were no organ weight changes, no macroscopic observations at necropsy and no histological changes considered to be related to treatment. There were some minor changes in hematology and clinical chemistry parameters, but these changes were small and considered to be of no biological or toxicological significance. There were no significant changes in plasma cholesterol levels as one may have expected from the consumption of AF.
