본 연구에서는 다양한 조건에서 돈분뇨를 처리하는 혐기소화 설비에서의 미생물 군집 구조를 심도 있게 연구하였다. 농장 규모 혐기소화 설비 Start-up 시 독성 물질에 대한 저항성이 높은 수소 이용성메탄균 우점화 연구를 수행하였으며, 돈분뇨를 처리하고 있는 국내의 실규모 설비들을 대상으로, 454 pyrosequencing 을 수행하여 핵심적인 박테리아와 메탄균 군집을 조사하고 이를 고찰하였다. 문헌에 근거하여, 초기 시운전 시에는 중온 조건으로 설비를 운전하였다. 그러나 예상과는 달리 아세트산 이용성 메탄균 (MSL) 이 급격하게 우점화 되었으며, 이는 초기 슬러지 접종에 의한 낮은 암모니아 농도 때문으로 사료된다. 박테리아 군집의 경우, 특이한 변화는 없었다. 이 후 구제역이 발발하여 기질 투입을 189일간 중단하였다. 구제역 완화 이후, stage I 에서 아세트산 이용성 메탄균이 우점하였기 때문에, Stage II 에서는 수소 이용성 메탄균의 우점화를 달성시키기 위해, 고온 조건 (50°C) 으로 설비를 운전하였다. 고온 운전 기간시점에서는 MBT 와 MMB 로 대변되는 수소 이용성 메탄균이 98% 이상 우점화되었다. 특히, MMB 에 속하는 고온성 메탄균인 Methanoculleus receptaculi 가 새롭게 출현하였으며, 모든 중온성 메탄균의DGGE band intensity 가 감소하였다. 다변량 분석 결과는 HRT 의 경우, 모든 메탄균에 대해서 부정적인 상관관계가 있고, 유입수 pH 의 경우, 모든 메탄균 군집과 부정적인 상관관계가 있는 것으로 도출되었다. 박테리아 군집의 경우, 고온성 환경에서 isolation 된 미생물 군집들의 DGGE band intensity 가 증가하였다. Stage II 에서 수소 이용성 메탄균 우점화를 확인한 후, stage III 에서는 다시 중온 조건 (35°C) 으로 설비를 운전하였다. stage I 의 시운전 기간의 중온 운전 시점과는 달리stage III 에서는 수소 이용성 메탄균의 단일 우점 현상이 관찰되었다. 다변량 분석 결과, HRT 는 MMB 와는 긍정적 상관관계가 있으나, MBT 와는 부정적 상관관계가 있는 것으로 도출되었다. Stage III 에서는 새로운 중온성 syntrophic 박테리아 (C. ultunense and S. wolfei subsp. saponavida) 의 DGGE band intensity 가 증가하였고, 이는 수소 이용성 메탄균의 우점화 현상이 기여했다고 사료된다. 결론적으로, 운전 온도 조절에 따라, 혐기소화 설비 start-up 시 수소 이용성 메탄균 군집을 성공적으로 우점화시켰으며, 유리 암모니아가 인자라고 사료된다. 또한 syntrophic 박테리아 또한 수소이용성 메탄균 우점화 현상에 기여했을 것으로 사료된다. 아울러, 454 pyrosequencing 방법을 사용하여 돈분뇨를 처리하는 여러 실규모 혐기소화조들의 미생물 군집 구조를 분석하고, 고찰하였다. pyrosequencing 분석을 통해 박테리아의 경우, 총 107,832 개, 고세균의 경우, 16,758 개의 염기서열을 얻었다. 또한, 박테리아의 경우 442 개의 OTU 로 구분되었으며, 고세균의 경우 42개의 OTU 으로 구분되었다. 홍성 혐기소화조 박테리아 군집의 경우, 71개의 OTU 로 구분되었으며, phylum 단위에서는 Proteobacteria (36.7%), Firmicutes (27.3%), Bacteroidetes (13.5%), Deinococcus-Thermus (8.0%), Spirochaetes (4.8%) 순으로 분석되었다. 핵심 genus 는 Pseudomonas, Arcobacter, Bacillus, Truepera, Fastidiosipila, Alkaliphilus 으로 도출되었다. 공주 혐기소화조 설비의 경우, 102개의 OTU 가 생성되었으며, phylum 수준에서는 Proteobacteria (62.1%), Firmicutes (19.9%), Bacteroidetes (9.9%), Spirochaetes (1.6%) 순으로 박테리아 군집이 조성되어 있었으며, Pseudomonas, Thauera, Geobacter, Bacillus, Tissierella, Fastidiosipila, Desulfitibacter 가 핵심 genus 으로 도출되었다. 순천 혐기소화조의 경우, 총 172 개의 OTU 가 생성되었으며, phylum 수준에서는 Firmicutes (54.0%), Tenericutes (21.1%), Bacteroidetes (14.3%), Proteobacteria (8.0%), Spirochaetes (6.8%) 으로 군집 구조를 형성하고 있었다. 핵심 genus 는 Acholeplasma, Alkaliphilus, Fastidiosipila, Truepera, Syntrophomonas 으로 도출되었다. 마지막으로 포천 혐기소화조의 박테리아 군집은 97 개의 OTU 으로 분류되었으며, phylum 수준에서는 Firmicutes (87.6%), Bacteroidetes (6.1%), Spirochaetes (1.3%) 으로 구성되어 있었다. 핵심 genus 는 Coprothermobacter, Fastidiosipila, Alkaliphilus, Gelria, Tepidimicrobium, Syntrophomonas 으로 도출되었다. 홍성 혐기소화조의 메탄균 군집은 10개의 OTU 으로 구분되었으며, order 수준에서MMB (70.2%), MSL (29.3%), MBT (0.5%) 으로 구성하고 있었다. 공주 혐기소화조의 메탄균 군집은 12개의 OTU 를 갖는 것으로 분석되었으며, MMB (75.2%), MSL (24.4%), MBT (0.5%) 순으로 존재하였다. 순천 혐기소화조의 메탄균 군집은 10개의 OTU 를 갖는 것으로 분석되었으며, MMB (93.2%), MSL (4.2%), MBT (2.6%) 순으로 존재하였다. 끝으로, 포천 혐기소화조의 메탄균 군집은 10개의 OTU 를 갖는 것으로 분석되었으며, MMB (56.6%), MBT (35.7%), MSL (4.7%) 순으로 존재하였다. 결론적으로 박테리아 군집의 경우, 소화조 설비에 따라 매우 다양하였으나, 일부 genus 들은 공통적으로 핵심적인 역할을 수행하고 있었다. Acholeplasma, Alkaliphilus, Fastidiosipila, Proteiniphilum, Pseudomonas, Tissierella 가 모든 혐기소화조에서 발견되었으며, Alkaliflexus, Clostridium, Petrimonas, Syntrophomonas, Truepera 는 3개의 혐기소화조에서 공통으로 발견되었다. 특히, Fastidiosipila 은 모든 소화조에서 높은 비율로 발견되었으며, Alkaliphilus, Pseudomonas, Bacillus 또한 돈분뇨 혐기소화조에서 우점하는 군집이었다. 메탄균의 경우, 수소 이용성 메탄균이 반응조 형태, 계절적 변화 등과 관계 없이 항상 우점하였다. 본 연구결과는 적절한 온도 조절을 통해 혐기소화조 start-up 수행 시, 독성 물질에 대해 강한 저항성을 갖고 있는 수소 이용성 메탄균의 효과적인 우점화 가능성을 제시하고 있다. 아울러, 454 pyrosequencing 메탄균 군집 결과는 핵심적인 박테리아와 수소 이용성 메탄균이 돈분뇨를 처리하는 혐기소화조 설비에서 우점하고 있음을 나타낸다. 이는 혐기소화 설비 식종원 탐색 시, 유용한 자료로 활용 될 수 있을 것으로 기대된다.

The microbial communities were examined at various scale anaerobic digestion (AD) plants treating swine wastewater. In the first chapter, the farm-scale AD process treating swine wastewater was operated over 591 days with changes of operating temperature and HRT and under unforeseen circumstances of starvation. Growth of various species of mesophilic AMs and HMs increased in the pre-operation period. However, the AMs, MSL, were the most dominant methanogenic group among the three methanogenic orders at the end of pre-operation period. The shifts in the methanogenic community structure in this are mainly caused by the operating temperature itself, 35°C and relatively low ammonia concentration. As previously mentioned, the AD process was should be stooped for 189 days due to FMD. MBT population was maintained during the long starvation rather than other methanogens. There was no obvious community shift of bacteria in stage I. After alleviation of FMD, the operating temperature for the AD system was adjusted to thermophilic, 50°C. HMs were successfully stabilized as a dominant group of methanogens in the AD system at stage II. More than 98% of the total methanogenic population was occupied by HMs, consisting of MBT and MMB. Especially, Methanoculleus receptaculi, thermophilic microorganism, belonging to MMB that gradually appeared as time elapsed. All identified mesophilic methanogens have been disappeared during stage II. HRT negatively correlated with all three methanogenic orders, indicating 50 days HRT was a relatively more suitable condition for methanogenic growth at HRTs longer than 100 days. PH of influent, which varied between 7.0 and 8.1, was also negatively correlated with all three methanogenic orders. In case of bacterial community, the band intensity of thermophilic bacterial clone increased stage II. In stage III, the operating temperature was adjusted to mesophilic conditions, 35 °C, again because the domination of HMs was already confirmed in the previous stage. HMs were still dominated during stage III despite of mesophilic operation. In contrast to stage I when the populations of AMs and HMs increased together, only the population of HMs increased and became dominant during stage III. The order MMB is the most dominant methanogenic group in the period of HRT of 50 days. The population of M. bourgensis increased and its maximum population was 3.8 x 107 copies/mL on Day 421. The RDA results indicate that HRT had a positive correlation with MMB and a negative correlation with MBT. Another syntrophic partner of HMs was newly identified by Clostridium ultunense and Syntrophomonas wolfei subsp. saponavida in the stage III. The dominance of HMs must be encouraged by these syntrophic microbial groups. The highest number of sequence reads were detected within Proteobacteria (36.7%), followed by those within Firmicutes (27.3%), Bacteroidetes (13.5%), Deinococcus-Thermus (8.0%), and Spirochaetes (4.8%). The major genus was detected by Pseudomonas, Arcobacter, Bacillus, Truepera, Fastidiosipila, and Alkaliphilus, respectively. Taken together, the bacterial communities were greatly diverse according to the anaerobic digesters treating swine wastewater. Although it was difficult to evaluate rules in microbial communities, some genus could be regarded as core microbial communities in the AD process. Six genus including Acholeplasma, Alkaliphilus, Fastidiosipila, Proteiniphilum, Pseudomonas, and Tissierella were ubiquitously detected at all the full-scale AD process. In addition, five genus including Alkaliflexus, Clostridium, Petrimonas, Syntrophomonas, Truepera were detected in the three AD process. Especially, the Fastidiosipila, the microbial group of protein degrader, was ubiquitously existed in all anaerobic digester treating swine wastewater with highly relative composition. The Alkaliphilus, Pseudomonas, and Bacillus were also the dominated in the AD process treating swine wastewater. HMs was absolutely dominant methanogenic microbial group in the AD process treating swine wastewater. FA was considered as the major factor for inducing predominance of HMs in AD process from the results. It could be conclude that HMs most likely play an important role in methanogenesis in AD process treating swine wastewater regardless of reactor types, seasonal variation and the substrate characteristics.

The major genus was detected by Pseudomonas, Arcobacter, Bacillus, Truepera, Fastidiosipila, and Alkaliphilus, respectively. A total of 10 OTUs was generated in the ADSC over the whole samples. Finally, a total of 10 OTUs was generated in the AD process in the ADPC over the whole samples. Taken together, the bacterial communities were greatly diverse according to the anaerobic digesters treating swine wastewater. Although it was difficult to evaluate rules in microbial communities, some genus could be regarded as core microbial communities in the AD process. Six genus including Acholeplasma, Alkaliphilus, Fastidiosipila, Proteiniphilum, Pseudomonas, and Tissierella were ubiquitously detected at all the full-scale AD process. In addition, five genus including Alkaliflexus, Clostridium, Petrimonas, Syntrophomonas, Truepera were detected in the three AD process. Especially, the Fastidiosipila, the microbial group of protein degrader, was ubiquitously existed in all anaerobic digester treating swine wastewater with highly relative composition. The Alkaliphilus, Pseudomonas, and Bacillus were also the dominated in the AD process treating swine wastewater. HMs was absolutely dominant methanogenic microbial group in the AD process treating swine wastewater. FA was considered as the major factor for inducing predominance of HMs in AD process from the results. It could be conclude that HMs most likely play an important role in methanogenesis in AD process treating swine wastewater regardless of reactor types, seasonal variation and the substrate characteristics.
