The senescent WI38 cells were permanently arrested and exhibited a senescent phenotype including enlarged and flattened cell morphology and increased senescence associated β-galactosidase (SA-β-gal) activity. The induction of WI38 cells senescence was associated with an activation of p53, increased expression of p21Waf1/Cip1 and p16Ink4a, and hypophosphorylation of retinoblastoma (Rb) protein were observed. In this study, I found that daidzein considerably reversed senescence-like phenotypes in the H2O2 induced premature senescence, including alterations of morphology, SA-β-gal staining, as well as related molecules expressions, p16Ink4a and P-Rb, while no changes in the expression of p53 and p21Waf1/Cip1. Daidzein was also found to reduce the expression of P-S6 (mTOR activity) during H2O2 induced premature senescence. And H2O2-induced SA-β-gal positive cells were reduced by rapamycin (a specific inhibitor of mTOR) in dose-dependently. Thus, I demonstrated that daidzein protected H2O2-induced cellular senescence via inhibition of p16Ink4a/Rb pathway and mTOR pathway in WI38 cells.
