The purpose of the study was to investigate the effects of 12 week vitamin D supplementation and circuit training on skeletal muscle mass and protein synthesis signals in vitamin D deficient type 2 diabetes women. Forty-eight vitamin D deficient type 2 diabetes elderly women, aged 63-84 years, were randomly assigned to control group(n=10), vitamin D group(n=11), exercise group(n=12), and vitamin D+exercise group(n=15). Vitamin D group and vitamin D+exercise group received 1,200 IU of vitamin D orally a day, and for placebo effect, control group and exercise group received placebo tablets. Exercise group and vitamin D+exercise group participated in circuit training, performed at 60-80% of HRR, 45-60 minutes a day, 3-4 days a week for 12 weeks. Body composition parameters were measured by Dual Energy X-ray Absorptiometry. Blood samples were drawn in the morning after 12hour fast. 25(OH)D, fasting glucose, insulin, Akt, and mTOR were analyzed. All measurements were conducted at pre and post of 12 week intervention. After 12 weeks of intervention, body weight was decreased significantly in vitamin D(p<.05), exercise(p<.05), and vitamin D+exercise(p<.001) group. Body mass index and percent body fat were decreased in exercise(p<.05) and vitamin D+exercise group(p<.001, p<.01). An increase in serum 25(OH)D was greater in the vitamin D(p<.001) and vitamin D+exercise(p<.001) group than exercise group(p<.05). There was no significant change in fasting glucose, insulin, and HOMA-IR in all groups. Appendicular skeletal muscle mass and skeletal muscle mass index were significantly increased in only vitamin D+exercise group(p<.01). Protein synthesis signaling pathway indicators, Akt and mTOR, were significantly decreased in all groups.
