Most of the neutralizing antibodies of HA which counter viral infectivity and provide protection against infection are directed against the highly variable HA1 regions of HA protein. While the HA2 stalk domain is relatively well conserved and induced broadly neutralizing antibody across the subtypes. Therefore, in this study, we demonstrated the successful generation of novel chimeric recombinant viruses possessing the whole HA1 region of 2009 pH1N1(A/Korea/CJ/01/09) and the HA2 region from HPAI H5N1 (A/EM/Korea/w149/06)[group1] and the HA1 region of group 1 HA subtype (H1N1) and HA2 regions of group 2 (A/Perth/06, H3N2) using reverse genetics (RG) system. Experimental animal study results revealed that the HA2 stalk-specific antibody responses are induced by vaccination with these chimeric RG viruses. Further, the chimeric HA virus vaccination are sufficient to inhibit viral replication and protect immunized mice from lethal challenge of pH1N1, HPAI H5N1, and mouse-adapted H3N2 viruses.
