Currently, the coexistence of a 26S proteasome with HslVU protease in the same living organism in eukaryotes has been reported. Although, HslVU complexe is found in both prokaryotic and eukaryotic organisms, it has only been characterized in prokaryotes. However, it is elusive in eukaryotic counterpart. One HslV peptidase was present in each species of prokaryotes, archaea, and eukaryotes, and one HslU ATPase in prokaryotes, but two HslUs was found in the mitochonria of eukaryotes exist only. In Trypanosoma brucei, a protozoan parasite that causes human sleeping sickness in Africa, one HslV and two HslUs are located in the mitochondria, where it has a novel function in regulating mitochondrial DNA replication. Here, I report the crystal structure of a first eukaryotic HslV from Trypanosoma brucei, determined at 2.4 Å resolution. By a comparison for the biochemical and structural data, I discuss herein the differences and similarities between eukaryotic and prokaryotic HslVs. Furthermore, this study is the biochemical characteristics of an HslVU complex from T. brucei, I found that only one of HslUs activates HslV. A tyrosine residue at the C-terminal tail of HslU2 has been identified as a key determinant for activating HslV in T. brucei and a phenylalanine at the equivalent position is found in T. brucei HslU1 as E. coli HslU. Unexpectedly, I found the F441Y mutation in HslU enhanced the peptidase and caseinolytic activity of HslV in E. coli. However, it showed partially reduced ATPase and SulA degradation activity. To begin with, only the C-terminal tail of HslU has been focused on for the study of HslV activation mechanism, however, the interacting Pro315 residue with Phe441 in free HslU is also found as a critical residue for HslV activation. Thus, my biochemical data explore the importance of the loop re-gion right before Pro315 residue for functional HslVU complex. The proline and phenylalanine pair in prokaryotic HslU has been replaced with the threonine and tyrosine pair in the functional eukaryotic HslU2. The sequence comparison among many HslUs from different kingdoms in combination with biochemical activity results of E. coli mutants shed light on the molecular evolutionary pathway of HslUs.
