The purpose of this research was to optimize process factors of liposome preparation in order to improve encapsulation of lysozyme in liposomes and to investigate pharmaceutical properties of freeze-dried formulation of high efficiency liposomes for efficient and stable drug delivery. High efficiency liposomes of lysozyme were prepared by reverse phase evaporation method, and drug encapsulation of liposomes was studied for several process factors including organic solvents, lysozyme/PC weight ratio and water : organic phase volume ratio. Loading amount and loading efficiency of liposomes were highly enhanced when prepared with diethyl ether at lysozyme/PC weight ratio of 0.18 and water : organic phase volume ratio below 1:3. Based on these results, the high efficiency liposomes of lysozyme were obtained by optimized procedures and then freeze-dried with several cryoprotectants. Physical characteristics and formulation stability of reconstituted suspension of freeze-dried liposomes were evaluated, compared to them of primary liposomes without freeze-drying. As results, the liposomes freeze-dried with glucose and trehalose as cryoprotectants turned out to have good physical stability in terms of particle size, polydispersity index and zeta potential. Also, encapsulated lysozyme in dry state was examined by FT-IR analysis and the peaks assigned to characteristic peptide structures of lysozyme were observed in dry liposome formulations, indicating significant incorporation of protein drugs in dry liposomes. Consequently, it is expected that dry liposomal formulation, which highly incorporates lysozyme, would be utilized in various formulations for protein drug delivery including parenteral, oral, nasal, and pulmonary routes. And the effects of process factors on drug encapsulation could be considered at the stage of developing liposomal formulations of many hydrophilic protein therapeutics besides lysozyme.
