The combination of improved technology and non-invasive sampling has been proposed as a new approach in medical prognostics. In this study, we developed a screening-based method to distinguish between normal healthy persons and patients with Alzheimer’s (AD) and Parkinson’s diseases (PD) using saliva and exhaled breath. We collected 20 each human samples of saliva and exhaled breath from those AD and PD patients and also normal healthy persons. All the participants recruited were clinically diagnosed before being used in this study. Exhaled breath samples were analysed using breath sensor array, LUSH-based extended gate-ion-sensitive field-effect transistor (EG-ISFET) biosensor, and gas chromatography-mass spectrophotometry (GC-MS). Salivary trehalose was determined from those samples using cell-based ISFET and EG-ISFET biosensors. In addition to biosensors, salivary Aβ 42, total tau (t-tau), phosphorylated tau (p-tau), expression of AD related genes, and pH have also been carried out. Our results showed that chemical compounds in the exhaled breath of patients were varied from those normal persons. Breath sensor array could distinguish most of the normal individuals from patients. In addition, GC-MS results showed that PD patients had higher occurrence and area percentage of siloxane than others. Besides, 1,2 benzenedicarboxylic acid diethyl ester, 1-phenanthrenol, and ethyl 3-cyano-2,3 bis (2,5 dimethyl-3 thienyl)-acrylate were consistently found in higher area percentage in AD patients compared others. On the other hand, cell-based EG-ISFET sensor demonstrated that the change of current for human saliva of patients with AD were distinctively different from the PD patients and normal persons. This suggests that the content of sugar present in the saliva differed among them. We also found slightly higher levels of salivary p-tau in patients with AD compared to normal persons and PD patients. Expression of AD-related genes increased in those AD patients than the normal persons. In addition, the pH of normal persons was more alkaline than those patients. The findings of this study may suggest the potential and great prospects of saliva and exhaled breath as non-invasive specimen samples for disease prognostic in the near future.
