Three-dimensional network of carbon nanotubes (3DN CNTs) has a net like structure which enhances the surface area and their local topographic interactions with analyte in flow system for increasing the detection limits in a short detection time.
In this thesis, the surface of 3DN CNTs was modified by steps and ultimately coated with CA125 antibodies to fabricate new CA125 sensor platform integrated with the microfluidic chip for point-of-care application. The surface modification of 3DN CNTs that was synthesized on micro-pillar structure was investigated by steps with atomic force microscope (AFM), contact angle, scanning electron microscope (SEM) and fluorescence microscope. In streptavidin step, the strength of 3DN CNTs as a biosensor platform was studied by comparing 2D system, 3D pillar structure and 3DN CNTs. The detection of CA125 was performed, with 3DN CNTs by fluorescence method and compared between two systems, in a static and fluidic system.
The detection limit of streptavidin with 3DN CNTs was 100 times lower than that of 2D and 10 times lower than that of 3D pillar structure. The detection limit of 3DN CNTs in a static system was 25 U/mL which is comparable with a commercial ELISA kit. The detection time was decreased by one fourth, 30min, in a fluidic system while the detection limit was maintained.
