This study was conducted to establish an efficient screening system for resistant melon to Fusarium oxysporum f. sp. melonis. GR isolate was isolated from infected melon plants collected at Goryeong and identified as F. oxysporum f. sp. melonis based on the morphological characteristics, molecular analyses, and host-specificity test on cucurbits including melon, oriental melon, cucumber, and watermelon. In addition, the GR isolate was determined as race 1 by resistance response of melon differentials to the fungus. To select optimized medium for mass production of inoculum of F. oxysporum f. sp. melonis GR, six media were tested. In V8-juice broth, the fungus produced the most spores (microconidia). On the other hand, resistance degrees of 22 commercial melon cultivars and 6 rootstocks for melon plant to the GR isolate were investigated. All the tested rootstocks showed no symptom of Fusarium wilt. Among the tested melon cultivars, only three cultivars were susceptible and the other cultivars represented moderately to high resistance to the GR isolate. For further study, six melon cultivars (Redqueen, Summercool, Superseji, Asiapapaya, Eolukpapaya, and Asiahwanggeum) showing different degree of resistance to the fungus were selected. Also, the development of Fusarium wilt on the cultivars according to several conditions such as plant growth stage, root wounding, dipping period of roots in spore suspension, inoculum density, and incubation temperature to develop the disease was tested. On the basis of the test results, we suggest that an efficient screening method for resistant melon plants to F. oxysporum f. sp. melonis is to remove soil from roots of seven-day-old melon seedlings, to dip the seedlings without cutting by scissors in spore suspension of 3 × 105 conidia·mL-1 for 30 minutes, to transplant the inoculated seedlings to plastic pot with horticulture nursery media, and then to cultivate the plants in a growth room at 25 to 28℃ for about 3 weeks with 12-hour light a day.

However, the inoculation method is required a lot of labor and time consuming because of the complicating inoculation procedure. To develop simple screening method, occurrence of Fusarium wilt on six melon cultivars (three susceptible and three resistant cultivars) according to inoculation method including root dip, scalpel, tip and soil-drenching methods was tested. Scalpel and tip method showed clear resistant and susceptible responses. However, tip method represented more variable disease severities. In the case of soil-drenching method, the susceptible cultivars did not represented clearly the characteristics. Thus, we selected scalpel method as a simple inoculation method to resistant melon cultivars to Fusarium wilt. And resistance responses of the six cultivars according to incubation temperature and inoculum density were investigated by use of the scalpel inoculation method. Incubation temperatures (25 and 30℃) and inoculum density (1.0 × 106 and 1.0 × 107 conidia·mL-1) was hardly affect the resistance degree of the cultivars.
