그러므로 본 학위 논문의 목표는: (a) 첫 번째로 대장균에서 발현한 M2e (1xM2e or 4xM2e) 단백질과 혼합된 불활성화 H9N2 (a-H9N2) 백신의 범 아형 방어효능을 평가하는 것이며 (b) 두 번째 목표는 mouse-adapted A(H1N1)pdm09 바이러스 감염 실험을 통해 마우스와 페럿에서 최근 한방천연물 유래 물질인 KIOM-C의 항바이러스 효능검증을 확인하고자 하였다 (c) 마지막으로, 최근 백신효능 문제점의 하나인 B형 인플루엔자 바이러스의 증식성, 병원성 연구를 위해 포유류 숙주 적응성 B형 바이러스를 선별하였으며, 이를 이용하여 B형 바이러스가 병원성을 획득하는 기전 및 페럿에서의 전파 능에 관여하는 바이러스 인자를 규명 하였다.

Thus, seasonal influenza vaccines are prepared annually based on global epidemiological surveillance data. However, since there is no method by which to predict the influenza strain that will cause the next pandemic, and antiviral drugs are always concerned for mutations. The overall objectives of this thesis are: (a) to evaluate the efficacy of an inactivated H9N2 virus vaccine (a-H9N2) mixed with M2e (1xM2e or 4xM2e) proteins expressed in Escherichia coli in animal model; (b) to examine the efficacy of herbal antiviral compound (KIOM-C) against infection with a mouse-adapted A(H1N1)pdm09 virus, and; (c) to understand the molecular characterization through mammalian host adaptation of influenza B viruses.
The first, to initially evaluate the efficacy of an inactivated H9N2 virus vaccine (a-H9N2) mixed with M2e (1xM2e or 4xM2e) proteins expressed in Escherichia coli, which contains the consensus of sequence the extracellular domain of matrix 2 (M2e) of A/chicken/Vietnam/27262/09 (H5N1) avian influenza virus, vaccinated mice were challenged with lethal homologous (A/chicken /Korea/ma163/04, H9N2) or heterosubtypic virus (A/aquatic bird/Korea/maW81/05, H5N2 and A/Philippines/2/82, H3N2). Survival results demonstrated that mice immunized with a-H9N2/1xM2e or with a-H9N2/4xM2e was higher than that of mouse-adapted H9N2 virus-infected mice vaccinated with 1xM2e alone or with 4xM2e alone. Heterosubtypic (H5N2 and H3N2) virus survival results found that the a-H9N2/4×M2e group displayed cross-protection, whereas vaccine alone, 1×M2e alone, 4×M2e, or a-H9N2/1xM2e showed incomplete protection. These results suggest that prokaryote-expressed M2e protein improved inactivated H9N2 virus vaccine efficacy and achieved cross-protection against lethal influenza A virus infection in mice.

Additionally, oral administration of KIOM-C increased the production of antiviral cytokines, including IFN-γ and TNF-α , and decreased levels of pro-inflammatory cytokines (IL-6) and chemokines (KC, MCP-1) in the Bronchoalveolar lavage fluid (BALF) of A(H1N1)pdm-infected mice. In the third aim, B/Florida/04/2006 (B/Yamagata lineage) virus was serially passaged in mice until it caused highly virulence with 100% mortality and evaluated their molecular analysis. Biological properties of the mouse-adapted viruses were also investigated in ferret. Molecular analysis revealed that the five viruses; wild type (B/Florida/04/06, P0), 3 intermediate-passaged viruses (P5, P9, and P12), and the finally mouse-adapted virus (P17, MA) which showed complete lethality obtained one to five amino acid substitutions in four segments (HA, M, NP, and/or PA) compared to that of the parental (P0) virus. In viral growth kinetics of MDCK cells and in mice lungs, P17 (MA) virus enhanced replication property compared to the parental virus. Interestingly, P17 (MA) virus demonstrated increased viral titers in the upper respiratory tract of the infected ferrets and transmitted into naive direct-and aerosol indirect-contact ferrets in 3 dpc while no virus was recovered from any contact groups in another virus-infected ferrets. This study provides an insight on how influenza B viruses could adapt to a mammalian host through molecular evolutions that subsequently increase pathogenicity.
Collectively, the finding of this studies exhibited that M2e polymer protein (4X M2e) and potential herbal therapeutic compound (KIOM-C) would provide helpful options to prevent influenza A viruses.
