C57BL/6J 마우스를 8개의 그룹 즉, 정상식이군, MCD-diet 대조군, MCD+CLRE 200 mg/kg 처리군, MCD+CLR-Hx 200 mg/kg 처리군, MCD+CLR-MC 100 mg/kg 처리군, MCD+SIL 100 mg/kg 처리군, MCD+BDMC 100 mg/kg 처리군, MCD+SIL 50+BDMC 50 mg/kg 처리군 으로 나누어 4주간 사육 후 체중과 간 중량을 측정하고, 간 기능 검사와 조직 병리적 실험과 지방축적을 매개하는 매개체들의 발현을 실험하였다. OA로 유도한 HepG2 세포에 처리하여 세포내 지방생성을 Oil-red O 염색법과 효소측정법, Western blot 분석법, RT-PCR 분석법을 이용하여 측정 그 결과를 비교하였다.
그 결과, MCD-diet군에서는 mice의 체중 감소가 뚜렷이 나타났으나, MCD+CLRE 200 mg/kg 처리군, MCD+CLR-Hx 200 mg/kg 처리군, MCD+CLR-MC 100 mg/kg 처리군, MCD+SIL 100 mg/kg 처리군, MCD+BDMC 100 mg/kg 처리군, MCD+SIL 50+BDMC 50 mg/kg 처리군 에서는 MCD-diet군보다 체중감소가 심하지 않았으나 유의성은 인정되지 않았다. 간 중량 측정에서는 MCD-diet군에서는 간 중량이 감소하였으나, MCD+CLRE 200 mg/kg 처리군, MCD+CLR-Hx 200 mg/kg 처리군, MCD+CLR-MC 100 mg/kg 처리군, MCD+SIL 100 mg/kg 처리군, MCD+BDMC 100 mg/kg 처리군, MCD+SIL 50+BDMC 50 mg/kg 처리군 모두에서 mice의 간 중량이 증가 되었다. 혈중 중성지방 (TG), 총콜레스테롤 (TC), HDL-콜레스테롤, LDL-콜레스테롤 농도에서 유의적인 효과를 나타냈다. 또한, 조직병리학적인 실험에서 지방축적이 감소한 것을 확인할 수 있었고, 지방축적을 매개하는 매개체들 (SREBP-1c, FAS, PPAR-γ)의 발현을 농도 의존적으로 억제하였다. CLRE, CLR-Hx, CLR-MC 와 CUR, DMC, BDMC 처리군을 OA로 유도한 HepG2 세포에서 세포 내 지방축적과 TG, TC 생성을 농도 의존적으로 억제했다. 또한, 간의 지방축적을 매개하는 단백질 (SREBP-1, FAS, PPAR-γ) 발현에 유의적인 효과를 나타냈으며, AMPK 인산화를 증가시켰다.

After 4 weeks, body weight, liver weight, biochemical parameters for liver function test, histological changes, reverse transcription polymerase chain reaction (RT-PCR), and Western blot were assessed.
Hepatocytes were treated with OA containing with or without BDMC to observe the lipid accumulation. We also tested the effects of BDMC on triglycerides (TG) and total cholesterol (TC) in HepG2 cells. Western blot and RT-PCR was used to measure sterol regulatory element binding proteins-1 (SREBP-1), fatty acid synthase (FAS), peroxisome proliferator-activated receptor (PPAR)-α,γ and adenosine 5’-monophosphate (AMP)-activated protein kinase (AMPK) expression.

In lipid accumulation factors (such as SREBP-1c, FAS, PPAR-γ), MCD+CLRE 200 mg/kg group, MCD+CLR-Hx 200 mg/kg group, MCD+CLR-MC 100 mg/kg group, MCD+SIL 100 mg/kg group, MCD+SIL 50+BDMC 50 mg/kg group and MCD+BDMC 100 mg/kg group showed inhibitory effect on liver lipogenesis by reducing associated gene expressions caused by MCD diet.
In the HepG2 cells, BDMC suppressed OA-induced lipid accumulation and TG and TC levels. Also, BDMC decreased hepatic lipogenesis such as SREBP-1, FAS, and PPARγ. BDMC increased AMPK phosphorylation in hepatocytes.
In conclusion, our findings indicate that CLR and its active components has a potential suppressive effect on NAFLD. Therefore, our data suggest that CLR and its active components may act as a novel and potent therapeutic agent against NAFLD.
