이에 본 연구는 high throughput screening을 통하여 미생물 추출물과 식물 추출물의 항산화, 항염, 항균 활성을 탐색함과 동시에 후보 물질을 도출하여 효능을 검증 하였다. 스크리닝으로 부터MJM 10778, MJM 8637, MJM 2582 3 종의 미생물 추출물과 Artemisia capillaris, Adenophora triphylla 2종의 식물 추출물을 최종 선발 하였다.
제 1 장에서는 high throughput screening에 사용한 항산화, 항염 및 항균 활성의 탐색 방법과 미생물과 식물 추출물 라이브러리 제작 과정에 대하여 기술하였다.
제 2 장에서는 Streptomyces sp. MJM 10778의 균주 동정 및 항산화 활성을 규명 하였다. 균주는 높은 항산화 활성과 함께 낮은 세포독성을 나타낸 것을 확인하였으며, 산화적 손상으로 야기된 질병 치료에의 이용가능성을 제시하였다.
제 3 장에서는 Streptomyces sp. MJM 8637의 glutathione-S-transferase (GST) 억제 활성과 항염 활성을 규명하였다. 산림의 토양에서 분리한 MJM 8637균주는 계통학적 분석과 함께 GST-pi억제 활성, TNF-alpha 억제 활성을 가졌음을 확인하였으며, 낮은 세포 독성으로 인하여 다약재 내성 항암 치료에의 응용 가능성을 보여주었다.
제 4 장에서는 MJM 2582는 균주 분석을 통하여 Amycolatopsis 속의 새로운 종임을 동정하였다. 항균활성 물질은 항균 활성 측정을 기반으로 한 일련의 추출 및 분리정제 과정을 통하여 분리하였다. 구조 규명을 통하여 얻어진siderochelin A는 임상에서 분리한 항생제 내성 균주에 항균 활성이 있음을 확인 하였다.
제 5 장에서는 B16F10 melanoma cell을 이용한 Artemisia capillaris 추출물의 미백 활성 효능 및 작용기전을 이해하고자 하였으며 이는 멜라닌 합성 관련 단백질과 합성경로에의 작용을 억제시킴으로써 최종적으로 멜라닌 생성이 저해되었음을 확인하였다. 이러한 결과를 통하여 향후 과색소생성 및 기능성 화장품의 원료로서 Artemisia capillaris 추출물의 사용이 가능함을 확인하였다.
제 6 장에서는 Adenophora triphylla의 항비만 효과를 생체 내∙외의 실험을 통하여 검증하였다. 3T3-L1 지방세포와 동물 시험은 혈액학적, 조직학적, 세포 활성, 단백질 및 유전자 발현 실험을 으로 진행하여 항비만 효과를 검증하였다.
마지막으로, 제 7장에서는 high throughput screening을 통하여 탐색된 천연물의 생리 활성을 탐색함으로써 얻어진 본 연구에서 축적된 결과들이 질병의 치료와 예방 등의 기능성 원료로서의 활용 가능성이 있음을 기술하였다.

Medicinal developments has stimulated the research in the continuous study of new drugs or treatments in natural products sources that included plants, animals, and micro-organisms, because natural products were confirmed their safety and efficacy in history of traditional medicines. For example, ancestors applied to certain herbs or fermented organisms to relieve the pains, improve injuries, treat diseases, and healing.
Oxidative stress is a natural metabolic byproduct that occurs when oxygen molecules lose an electron and become into free radicals. Oxidative stress and inflammation are the basis of many age-related, chronic diseases.
In these days, natural products are big source of medicinal use that have taken a huge role in drug discovery and drug development. Moreover, that proceeded in step with science technology developments such as molecular biology, combinational chemistry, biotechnology. So, there were enormous efforts and investment of natural products to make a new advances. Now, natural products as a base of drug development and treatment of diseases, medicine and biology were combined their technology and knowledge. That allow to detail the exact biological effects of natural products on human.
In this study, I used three methods for High Throughput Screening include anti-oxidant activity assay, anti-inflammatory activity assay, and anti-bacterial assays with natural products, microorganism extracts and plants extracts to search for good candidates of natural products. Though this screening methods, I selected three microbial strains, MJM 10778, MJM 8637, MJM 2582 and two plants, Artemisia capillaris and Adenophora triphylla.
In the Chapter 2, I studied antioxidant activity of Streptomyces sp. MJM 10778 with performed 4 antioxidant tests and strain identification. This strain has a strong anti-oxidant activity and low cytotoxicity of the extract gives a possibility that the clinical use of the extract as a therapeutic agent of diseases, which are caused by free oxygen radicals.
In the Chapter 3, I found Streptomyces sp. MJM 8637 has glutathione-S-transferase inhibition and anti-inflammation activity. That was confirmed with phylogenic analysis and GST-pi enzyme assay, and TNF-alpha inhibition assay. Putatively novel, isolated from mountain forest soil and identified strain MJM 8637 has strong GST-pi inhibition and anti-TNF-α production activity. Moreover, the low cytotoxicity of the extract gives a possibility to develop the extract to an anti-cancer agent that can disable the multidrug resistance of cancer cells.
In the Chapter 4, I identified MJM 2582 is a new species of Amycolatopsis and isolation and purification of the active compound with several steps including organic chemical extraction, HPLC, and anti-microbial assays. The structure of purified compound was identidfied as siderochelin A and test its anti-microbial activity with clinical isolated microbes.
In the Chapter 5, I studied anti-melanogenesis mechanism of Artemisia capillaris extracts. From the results, Artemisia capillaris extracts markedly inhibits melanogenesis related proteins and regulated melanin synthesis pathway to lead inhibition of melanin production. So, I concludes Artemisia capillaris may be a good candidate to modulate melanogenesis in treatments for hyperpigmentation and cosmeceutical agents.
In the Chapter 6, I identified Adenophora triphylla was a food candidate for anti-obesity with in vitro and in vivo studies. That was consist with animal study, plasma and hepatic biological analysis, histological analysis, cell assay and western blot analysis.
This thesis study could help to understand the natural products from HTS with natural products, and individual studied products could be applied to improve disease treatment and care. The results are summarized in the Chapter 7.
