Many kinds of normal flora exist in the vagina of a female and lactobacilli of aerobic bacteria may account for the large percentage among these bacteria. The bacteria in vagina maintain a weak acid condition by the secretion of lactic acid and maintain the balance between vaginal microorganisms to get the resistance to pathogens, the chance of having infections increased by the situation resulting from change in the normal flora. There are various type of microorganism and virus causing inflammation due to vaginal infection, but most frequently observed vaginitis pathogens are trichomonas vaginitis, vaginitis caused by candida yeast infection, vaginitis caused by Actinomycetes due to intrauterine contraceptive device (IUD) use, and Gardnerella vaginalis, and these are the major vaginitis pathogens commonly observed within the vagina. In this study, three major vaginitis pathogens such as Gardnerella vaginalis, Candida albicans, Actinomycetes causing frequent vaginitis, their morphological characteristics were confirmed in the light microscope (LM) by Pap staining method, and examined the microstructure in the transmission electron microscopy (TEM) and scanning electron microscope (SEM). First, Pap staining method within cervico-vaginal squamous epithelium accompanied by trichomonas and bacterial vaginitis, Gardnerella vaginalis infected clue cells was identified. In the light microscope(LM) by Pap staining method, lactic acid bacteria maintaining acidic pH within vagina was not observed but some inflammatory cell was found. In the scanning electron microscope (SEM), Gardnerella vaginalis covering the cervico-vaginal squamous cell surface was produced and its growth environment was formed by the biofilm. These biofilm materials are either smoothly covered bacteria surface or connected between bacteria, and coccobacillus form with a various size about 0.3 - 0.6 um in diameter. The cell wall surface was coated with the small protruding substance, and some bacteria was confirmed the presence of breakup phase. Cervico-vaginal squamous cell cytoplasm infected with Gardnerella vaginalis in the transmission electron microscope (TEM) was filled with keratin fiber, and did not find any characteristics of cell organelles. Second, Candida albicans was confirmed as the vaginitis pathogens in the Pap staining method. Candida albicans observed in the light microscope (LM) with Pap staining method was formed the mass with vaginal epithelial cells. In the scanning electron microscope (SEM), Candida albicans cells were formed a community by producing hyphae, pseudohyphae and yeast, filamentous hyphae was elongated, and yeast cell was well observed. Budding phenomenon to form a new yeast was observed as a spherical or oval shape at the end of the yeast cell, and the yeast cell that form a daughter cells were smoothly covered with the biofilm materials. In the high magnification transmission electron microscopy (TEM), yeast cell was measured 3 - 3.5 um in a diameter, and biofilm surrounding the cell wall was measured about 400nm irregularly coated with fine fibrous materials. The diameter of yeast cell was observed 6 - 7 times larger than surrounding bacteria, and characteristically attached to the surface of inactive epithelial cells, and also confirmed that the yeast cell was existed as penetrating into the cytoplasm. Third, in a Pap staining method, Actinomycosis caused by Actinomycetes infection was observed around cervico-vaginal squamous cell with prolonged use of IUD. In the high magnification light microscope, Actinomycetes was attached to the surface of the cervico-vaginal squamous cell, or formed a colony by produce mass with inflammatory cells. The edge of the hyphae was observed like flock of wool, and thin filaments were showed as a protruding shape in the middle of flock. In the scanning electron microscope (SEM), surface of hyphae was coated with biofilm secreted from the bacteria, hexagonal sulfur granular materials were scattered around hyphae. These granular materials were accumulated together in a size about 100 to 150 nm in a diameter, and scattered around the Actinomycetes were observed. Hexagonal granular materials were analyzed with the Energy disperse X-ray spectroscopy (EDX) and a carbon (C), nitrogen (N), and sodium (Na), sulfur (S) was detected. Contrary to expectations that a large amount of sulfur (S) component would be detected, sulfur identified was accounted for 1.21%. Colony of Actinomycetes on the transmission electron microscope (TEM) was occupied an area of about 30 um2, cross-section of the hyphae and spores cutting at a various angles was shown in different size, high electron density granules were dispersed on the hyphael cytoplasm, and the diameter of these granules were about 50nm in a diameter and spherical shape was confirmed.
