The vestibulo-ocular reflex (VOR) refers to the compensatory eye movements that occur to maintain retinal images. The eyes automatically move in the opposite direction to that of head movement at almost an identical speed as that for the head movement. VOR indices, such as gain, phase, and symmetry are typically used to diagnose vestibular dysfunction. Peripheral vestibular signals affect body balance through the vestibulospinal reflex (VSR). Peripheral vestibular signals are known for their instantaneous control of the human body through vestibular nuclei in the brain stem and, their influence on spatial cognition through the cerebral cortex. Transcranial direct current stimulation (tDCS) is a non-invasive brain stimulation (NIBS) method that modulates neural excitation and inhibition of the cortical areas under the electrodes. Recently, tDCS on the temporoparietal junction (TPJ) has been shown to affect VOR, and tDCS on the motor cortex has been shown to enhance postural stability by reducing changes in the center of pressure. However, these previous studies have applied different current intensities, stimulation durations, and target areas. This study investigated a hypothesis that tDCS on the TPJ would enhance VOR indices and postural stability when high-intensity currents are implemented over a longer period of stimulation time. In the section on the Development of the VOR measurement system, we have listed the parts required for the low-cost multi-purpose VOR measurement hardware and discussed how to assemble it, with details of the structure and function of the VOR measurement software. The hardware consists of affordable parts that can be purchased easily. It was assembled based on a goggle-shaped main frame segment by attaching an accelerometer/gyroscope module and an infrared camera with near-infrared lightings. The software was designed with LabVIEW, a graphical programing language. It is possible to collect data, including the coordinates of the center of eye, from the infrared video and the accelerometer/gyroscope signals through Arduino, a microcontroller. In the section on the Performance test for the VOR measurement system, we have described horizontal VOR (hVOR) and vertical VOR (vVOR) measurements obtained using the custom-made VOR measurement system An autorotation test guided by visual and auditory cues revealed normal VOR in four healthy participants. However, alpha indices, which correspond to gain, approximated 1 only for hVOR in the small head rotation condition (10°). Despite the shortcomings of the present system, the results of performance test sufficiently demonstrate its utility as a psychophysiological measurement apparatus. In the section on the Experiment, we observed the effects of tDCS on hVOR and postural stability. By combining two levels of current intensities (1.0 mA or 2.0 mA) and two stimulation durations (10 min or 20 min), four verum stimulation conditions were set. In the sham-control condition, a 2.0 mA stimulation was delivered for 15 s. All participants were randomly assigned to five groups. To analyze the VOR change, this study used beta indices that represent the morphological integrity of VOR curves, as well as gamma indices that represent the phase difference in time units. To analyze postural stability changes, gyroscope data were divided into no movement (NM), low velocity movement (LVM), and high velocity movement (HVM). Their relative ratios were then calculated. The result showed no statistical differences in the effects of the tDCS parameters on VOR indices. However, tDCS affected postural stability indices. Especially, 2.0 mA/10 min condition exhibited a significant increase in the NM ratio and significant decreases of LVM ratio and HVM ratio. In conclusion, the 2.0 mA/10 min condition was more effective for maintaining postural stability than other conditions. And the result suggests that the vestibular cortex such as TPJ has more roles other than the modulation of VOR. The present study has several limitations. First, the custom-made VOR measurement system had a number of issues in its hardware and software. Therefore, the system requires technical improvements. Moreover, the system should be compared to commercial VOR measurement equipment. Second, the experiment was conducted using a between-participant design. In order to reduce any interference effects from individual differences, a within-participant design will be considered in a future study. Despite these shortcomings, the VOR measurement system described herein is expected to be utilized in future studies on the vestibular system because it can yield psychophysiological measurements. In addition, the observed effects of tDCS on postural stability suggests the usefulness of implementing tDCS for mitigating the symptoms of cybersickness.
