Abstract
Objectives : This study was performed to comparative investigate the distribution of primary sensory and motor neurons associated with “Daereung(PC7)”, “Cheonji(PC1)”, “Yangji(TE4)”, “Cheollyo(TE15)” acupoints by using neural tracing technique. Material & Methods : A total 16 SD rats were used in the present study. After anesthesia, the rats received microinjection of 6㎕ of cholera toxin B subunit(CTB) into the corresponding sites of the acupoints Daereung(PC7)”, “Cheonji(PC1)”, “Yangji(TE4)”, “Cheollyo(TE15)” in the human body for observing the distribution of the related primary sensory neurons in dorsal root ganglia(DRGs) and motor neurons in the spinal cord(C3∼T4) and sympathetic ganglia. Three days after the microinjection, the rats were anesthetized and transcardially perfused saline and 4% paraformaldehyde, followed by routine section of the DRGs, sympathetic chain ganglia(SCGs) and spinal cord. Labeled neurons and nerve fibers were detected by immunohistochemical method and observed by light microscope equipped with a digital camera. The labeled neurons were recorded and counted. Results : The CTB labeled areas projecting to Daereung(PC7) were observed in C5∼T1 spinal cord segments, C5∼T2 DRG and MCG, STG and T4 sympathetic ganglia. Double stained calcitonin gene related peptide(CGRP) and substance P(SP) fibers were observed in lamina III-IV areas of T1 or C8 spinal cord segments. Double stained CGRP and SP neurons were observed same level of DRG. The CTB labeled areas projecting to Cheonji(PC1) were observed in C6∼T4 spinal cord segments, C6∼T4 DRG and MCG, STG and T4∼T6 sympathetic ganglia. Double stained CGRP and SP fibers were observed in lamina I-II areas of C8 spinal segment. Double stained CGRP and SP neurons were observed same level of DRG. The CTB labeled areas projecting to Yangji(TE4) were observed in C5∼T2 spinal cord segments, C5∼T2 DRG and MCG, STG and T4 sympathetic ganglia. Double stained CGRP and SP fibers were observed in lamina I-III areas of C8 spinal segment. Double stained CGRP and SP neurons were observed same level of DRG. The CTB labeled areas projecting to Cheollyo(TE15) were observed in C3∼T1 spinal cord segments, C3∼T1 DRG and SCG, MCG, STG and T4∼6 sympathetic ganglia. Double stained CGRP and SP fibers were observed in lamina I-III areas of C3 spinal segment. Double stained CGRP and SP neurons were observed same level of DRG. Conclusion : From this research, the distribution of primary sensory and motor neurons associated with “Daereung(PC7)”, “Cheonji(PC1)”, “Yangji(TE4)”, “Cheollyo(TE15)” acupoints were concluded as follows. Muscle meridian related Daereung(PC7) and Cheonji(PC1) are controlled by spinal segments of C5∼T1, C6∼T4, respectively. As well as, muscle meridian related Yangji(TE4) and Cheollyo(TE15) are controlled by spinal segments of C5∼T2, C3∼T1, respectively.
