BACE2 is a membrane-bound aspartic protease highly homologous with BACE1. While BACE1 processes the amyloid precursor protein (APP) at a key step in generating the b-amyloid peptide and presumably causes Alzheimer’s disease (AD), BACE2 has not been demonstrated to be directly involved in APP processing, and its physiological functions remain to be determined. To determination of the original function and development of inhibitors from marine sources, we have constructed overexpression vector for the production of the BACE2. The gene encoding human BACE2 protease was amplified using polymerase chain reaction and cloned into the pET11a expression vector, resulting in pET11a/BACE2. Recombinant BACE2 protease was successfully overexpressed in E. coli as inclusion bodies, refolded by using the rapid-dilution method, and purified by two-step fast protein liquid chromatography using Sephacryl S-300 gel filtration and Resource-Q column chromatographies. In addition, the produced BACE2 protease was found to be an active form. This study provides efficient methods not only for the study of basic properties of the BACE2, but also for the development of inhibitors from marine natural sources.
