Influenza viruses are divided into A, B, and C types and the seasonal flu is usually caused by influenza A. Influenza viruses cause pandemic by antigenic shift and drift. In the parts I and II of this thesis, I isolated aptamers that showed an inhibitory effect on NS1 protein of influenza virus. The innate immune response starts by binding of RIG-I to viral dsRNA. Then, TRIM25 protein binds to activated RIG-I and mediates the RIG-I ubiquitination. The MAVS of mitochondria is activated by open form of RIG-I, which causes the expression of IFN-β. At this time, the viral protein NS1 inhibits the ubiquitination by TRIM25 and innate immune response is blocked. Therefore, isolation of novel aptamers that have an inhibitory effect on the NS1 is a good strategy to suppress the viral replication. In this study, I performed SELEX experiments to isolate the aptamers with DNA and RNA libraries and measured the affinity by ELISA. Then, TA cloning was performed with the pool that had the highest affinity. Finally, I was able to select one ssDNA aptamer and two RNA aptamers (R-1, R-2). ELISA and SPR experiments showed that the selected aptamers had nanomolar range of Kd values. Confocal and FACS analysis were performed in order to show the binding of aptamer to NS1 in mammalian cell. To determine the inhibitory effect of the selected aptamers on NS1, I performed luciferase assay, real-time PCR and ELISA, respectively. I also showed that the aptamers were able to suppress the replication of the influenza virus. Interestingly, pull down assay showed that two RNA aptamers were able to inhibit the interaction of NS1 with TRIM25. The recovery of TRIM25 activity was confirmed through ubiquitination assay. Therefore, the selected in this study would be utilized as therapeutic agents that could inhibit the replication of influenza virus. In the part 3 and 4 of this thesis, I isolated aptamers that specifically bind to hemagglutinin of influenza virus for diagnostic purpose. The counter-SELEX was performed to isolate the aptamers that could distinguish between the H1-HA1 and H5-HA1. Three aptamers, which showed the highest affinity to H1-HA1, were selected after 14 rounds of the counter-SELEX procedure. The selected aptamers were able to distinguish H1-HA1 from H5-HA1 and showed nanomolar range of Kd values by ELISA, SPR, and Western blot. Further experiments were performed to test the possibility of the selected aptamers as the probe of diagnostic sensor. After the immobilization of the aptamers on the silicon wafer, ELISA and FE-SEM were performed with HA1 protein. The contact angle of each step was measured and filmed. The electrical current changes by the binding of HA1 with immobilized aptamers on silicon wafer were also measured through CV. Based on this study, It is expected that the selected aptamers can be further developed as a useful probe for diagnosis of various subtypes of influenza virus.
