Tepidimicrobium ferriphilum can survive in an alkaline environment of pH 9 while using Fe (III) as a final electron acceptor. It is known as an extremophile because it can also withstand 5.4 kGay gamma ray exposure. In an anaerobic environment, iron-reduction process processes have important implications for metal phosphate transformation and organics decomposition. Therefore, the extremophile Tepidimicrobium ferriphilum might have various applications to diverse fields of industry. For the purpose of understanding ecological functions of the extremophile in soil environments, occurrence of the species in various soils were investigated. Because its cultivation is time consuming and the procedures are elaborated, species-specific primer targeting 16S rRNA gene of Tepidimicrobium ferriphilum was developed. By applying the PCR method, 14 soils from diverse sources in Korea were subjected for occurrence of the species. Surprisingly, all 14 soils were positive for the PCR. Anaerobic cultivation using the samples resulted novel strains of the species. By searching MG-RAST database, 7.6% of soil metagenomes were revealed to contain the species. The metagenomes were from highly diverse environments. In conclusion, Tepidimicrobium ferriphilum seems have an ecological niche in soil environment as a member of normal flora although its physiology is that of an extremophile.
