Ferric thiocyanate method에 의한 지질과산화 억제능에서 흡광도의 변화를 측정한 결과, 분지아미노산 중 발린 100 mM에서 항산화능이 가장 높은 72.05%로 나타났다. 류신 100 mM의 경우, sodium nitroprusside assay에 의한 NO 소거능 측정에서 98.52 μM만큼 가장 적은 양의 NO를 생성하여 NO 소거능이 가장 뛰어남을 알 수 있었다. 둘째, 분지아미노산의 항암 효과를 검증하였다. 대장암 유래 세포인 HT-29 세포에 대하여 류신 50 mM과 100 mM이 각각 53.4±3.56%와68.8±2.9%의 세포독성 효과를 보여 분지아미노산 중 가장 뛰어난 효과를 보였다. Comet assay를 이용하여 백혈구 DNA 손상 억제 효과를 검증한 결과, 류신, 발린, 이소류신 각각 33.2±1.8, 30.1±0.9, 32.3±1.5 μg/mL의 IC50값을 나타내어 3종의 분지아미노산 모두 암 발생의 전단계인 세포 내 DNA 손상에 대한 보호 효과가 있음을 확인하였다. 셋째, 분지아미노산의 항염증 효과를 측정하였다. 대식세포의 NO 생성에 분지아미노산이 미치는 영향을 검증한 결과, 류신 100 mM의 경우, 81.15%만큼 NO 생성을 억제하여 가장 뛰어난 저해능을 나타냈으며, 발린과 이소류신은 각각 29.65%와 42.95%만큼의 저해능을 보였다. 세포 내 염증인자인 NO를 생산하는 효소인 iNOS의 유전자 발현 정도에 분지아미노산이 미치는 영향을 측정한 결과, 발린, 이소류신, 류신이 각각 47.31%, 23.66%, 89.61%의 발현 저해율을 나타냈다. 또한, 항염증관련 유전자의 발현에 분지아미노산이 미치는 영향을 측정한 결과, IL-6, COX-2의 발현이 모든 분지아미노산의 처리에 의해 감소하여 분지아미노산이 항염증 관련 사이토카인의 발현을 억제함을 알 수 있었다. 본 연구를 통해 분지아미노산의 항산화, 항암, 항염증 효과를 검증함으로써 식품, 사료 및 제약산업에서 건강기능성 소재로서 사용되기 위한 기초 자료로 활용되리라 사료된다.

They are commonly used as nutritional supplements; however, little is known about their health functional activities. The objective of this study was to evaluate the antioxidant, anti-cancer, and anti-inflammatory activities of BCAAs. The antioxidant activity of BCAAs were determined by measuring the inhibitory effect on lipid peroxidation and their nitric oxide (NO) scavenging activity. The inhibitory effect of BCAAs on lipid peroxidation was measured by the ferric thiocyanate (FTC) method, and their NO scavenging activity was evaluated using the sodium nitroprusside (SNP) assay. The FTC method results indicated that valine was the most effective of the BCAAs, with 100 mM of valine showing 72.05% antioxidative activity. An SNP assay conducted for 24 h produced the least amount of NO (98.52 μM) when it included 100 mM leucine compared to the other BCAAs (control: 132.20 μM). The impact of BCAAs on the anticancer activity was evaluated with the cytotoxic and anti-genotoxic activities in human cancer cell lines and human leukocytes. Leucine exhibited the highest cytotoxic activity against cancer cell lines. Against HT-29 cells, 53.4±3.56% and 68.8±2.9% cytotoxicity at 50 and 100 mM was measured. Isoleucine exhibited greater than 50% cytotoxicity against HT-29 and LoVo cells. Anti-genotoxic activities of BCAAs in human leukocytes were assessed using the alkaline comet assay. Results were shown a 50% reduction in DNA damage at 33.2±1.8, 30.1±0.9, and 32.3±1.5 μg/mL for leucine, valine, and isoleucine, respectively. The anti-inflammatory property of branched-chain amino acids (BCCAs) in suppressing the LPS-induced production of nitric oxide (NO) and cytokines was investigated. The NO assay was used to measure the inhibitory effect of BCAAs on NO production in LPS-stimulated RAW 264.7 macrophages. BCAAs inhibited LPS-induced NO production, with 100 mM leucine having the most pronounced effect, suppressing NO production by 81.15%. Valine and isoleucine also reduced NO production by 29.65% and 42.95%, respectively. Consistent with these results, BCAAs also suppressed the expression of iNOS, the enzyme that produces NO. iNOS expression was reduced by 47.31%, 23.66%, and 89.61% in LPS-treated cells supplemented with valine, isoleucine, and leucine, respectively. Additionally, all BCAAs decreased the production of the cytokines IL-6 and COX-2, indicating that they could inhibit pro-inflammatory cytokine production. Valine also inhibited the production of IL-1β and TNF-α. These findings suggested that BCAAs can be utilized to develop functional food as antioxidative, anticancer, and anti-inflammatory agents.
