SIRT1 유전자의 활성화나 발현 증가는 대사성 질환, 신경퇴행성 질환, 심혈관 질환과 같은 질병을 개선시키는 다양한 효능을 나타낸다고 알려져 있다. 동아시아 지역에서 자생하고 오래 전부터 건강식품으로 사용되어 온 약용 식물들의 검증을 통해서, 인삼열매의 에탄올 추출물이 SIRT1 유전자의 발현을 유도한다는 사실을 확인하였다. 본 연구에서는, 인삼열매 과육의 부탄올 분획에 대한 repeated column chromatography과 preparative HPLC 정제과정을 통해서 하나의 순수한 생리활성 물질을 얻을 수 있었다. 그것의 구조는 mass와 NMR 스펙트럼 데이터를 통해서 시링가레시놀(syringaresinol)로 확인되었다. 시링가레시놀 (4,4 -(1S,3aR,4S,6aR)-tetrahydro-1H,3H-furo[3,4-c]furan-1,4-diylbis(2,6-dimethoxyphenol))은 cAMP-PDE (cyclic AMP phosphodiesterase)에 대한 저해효과와 운동 시간을 늘려주는 효과와 같은 다양한 약리적 활성을 가지고 있는 리그난 종류이다. 시링가레시놀은 식물자체에서 하나의 우세한 광학이성질체로 존재하거나 두 가지 이성질체가 혼합되어있는 형태로 존재하고 있다. 광학이성질체의 비율의 차이는 리그난의 생물학적 활성에 영향을 줄 수 있기 때문에, 우리는 인삼열매 과육에서 분리된 시링가레시놀 광학이성질체 비율과 그것이 SIRT1 유전자 발현에 미치는 영향을 조사하였다. 입체화학적 분석을 통해서 인삼열매 과육에는 (+)-시링가레시놀이 우세하게 존재하고 있음을 확인하였고, 더불어 루시퍼라아제 리포터 분석과 크로마틴 면역침강 (chromatin immunoprecipitation, ChIP) 시험을 통해서 (-)-시링가레시놀이 아닌 (+)-시링가레시놀이 염기배열 특이적으로 SIRT1 promoter에 FOXO3 유전자의 결합을 유도하여 SIRT1 유전자의 발현을 유도하는 것을 확인하였다. FOXO3는 anti-aging signaling pathway의 가장 downstreamtarget 단백질로서 수명을 조절하게 된다. 본 연구에서는 (+)-시링가레시놀이 FOXO3에 직접 binding하여 그의 활성을 조절하는 것을 확인하였다. 즉, 시링가레시놀은 FOXO3을 활성화하여 그의 세포 내 위치를 핵으로 이동시키고, FOXO3의 target 유전자들의 발현을 조절하였다. 그리고 FOXO3 유전자는 시링가레시놀의 결합 부위를 가지고 있고 시링가레시놀이 노출되면 장수유전자를 유도하는 것을 확인하였다. 이러한 결과는 DAF-16이나 FOXO를 불활성화 시키면 시링가레시놀이 C. elegans의 수명을 연장시키지 못하였고 또한 Forkhead 도메인의 손상도 시링가레시놀과 FOXO3의 결합을 방해한다는 것도 확인하였다. 이러한, 시링가레시놀의 효과는 C. elegans의 수명을 연장시킬 수 있는 것으로 확인하였다. 본 연구결과는 인삼열매에서 분리한 (+)-시링가레시놀이 장수 물질로서 개발될 수 있음을 제안한다

As the interests in longevity and the possible components or drugs that may regulate longevity gene as well as metabolism have been increasing, a variety of pharmacological approaches have been executed. However, these efforts have been disturbed by the relative deficiency of identified molecular pathways with functionally druggable characters that can extend life span. The pharmacological activities of ginseng berry have also been reported such as anti-diabetic, anti-cancer, anti-oxidant, anti-aging and anti-stress. However, as ginseng berry is preserved for planting, it has not been generally used for medicines or health foods as much as the ginseng root. Several dietary compounds were screened to see if they are able to enhance the expression levels of sirtuin1 (SIRT1) gene. SIRT1, the mammalian orthologue of silent information regulator 2 (Sir2), is a nicotinamide (NAD+)-dependent deacetylases and plays diverse roles in a number of cellular processes through the deacetylation of key metabolic enzymes and transcriptional regulators, and the coordination of gene-expression programs. Activation or increased expression of SIRT1 has a broad spectrum of beneficial effects in metabolic diseases, neurodegenerative disorders and cardiovascular diseases. After extensive screening of the extracts from medicinal herbs that had traditionally been used as a health food for anti-aging in East Asia, the ethanol extract of ginseng berry pulp were confirmed to induce the expression level of SIRT1 gene. In this study, repeated column chromatography and preparative HPLC of butanol-soluble fraction of ginseng berry pulp led to one pure bioactive compound. Its structure was identified as syringaresinol via a comparison of its mass and NMR spectral data with those of authentic reference compound. Syringaresinol (4,4 -(1S,3aR,4S,6aR)-tetrahydro-1H,3H-furo[3,4-c]furan-1,4-diylbis(2,6-dimethoxyphenol)) is a lignan with several pharmacological activities; for instance, it has an inhibitory effect on cyclic AMP phosphodiesterase and a prolonging effect on exercise time. Syringaresinol exists either exclusively as one enantiomer or enantiomeric mixtures in plants. Given that the difference in enantiomeric ratio affects biological activities of lignans, the enantiomeric ratio of syringaresinol isolated from ginseng berry pulp and the enantioselective effects of syringaresinol on SIRT1 gene expression were investigated. Stereochemical analysis showed that Panax ginseng berry pulp contains predominantly the (+)-enantiomer of syringaresinol. Furthermore, using luciferase reporter assays and chromatin immunoprecipitation, it was verified that (+)-syringaresinol, but not (-)-syringaresinol, induced the binding of FOXO3 to the SIRT1 promoter in a sequence-specific manner, which lead to the induction of SIRT1 expression. FOXO3 is the most “downstream target protein” in the anti-aging signaling pathway and therefore controls the life span. In this study, (+)-syringaresinol regulated the activity of FOXO3 when it directly bound to FOXO3. In other words, syringaresinol activated FOXO and relocated itself within the cell to the nucleus which then regulated the expression of FOXO’s target genes. the FOXO3 gene contains a possible syringaresinol binding site and that the exposure of syringaresinol induces activation of the longevity gene. This idea is supported by the findings that dysfunction of DAF-16 or FOXO abrogates the effects of syringaresinol on life span and disruption of Forkhead domain prohibits the binding of syringaresinol with FOXO3. These data indicate that the effects of syringaresinol extended the life span of C. elegans. From these results, (+)-syringaresinol isolated from ginseng berry has the potential to be developed as a substance for longevity.
