This study investigated the distribution of pathogens that have the potential to cause human infections in the school areas where young students spend most of their time. Twenty-four places that students most commonly frequent were chosen for the study of the major pathogens. These included Staphylococcus aureus, including methicillin-resistant S. aureus (MRSA), which is increasingly isolated even in the community, and E. coli, which is a representative marker for fecal contamination. In addition, fungi were included in the study. Microorganisms were identified based on culture, microscopy, and 16S (for bacteria) or 18S (for fungi) ribosomal RNA (rRNA) gene sequence analyses. By the culture method, varied numbers of various forms of bacteria and fungi were isolated from most places. Places that were highly contaminated with microorganisms and less contaminated were determined. Based on the colony morphology characteristics on the culture media, the bacteria were able to be separated into seven major groups. The bacteria of each major group were further analyzed for the accurate identification of their species using 16S rRNA gene sequencing. The most frequently isolated bacteria were Staphylococcus species that probably originated from humans, and these were followed by tetracoccus forms that are common in the environment and then Gram-negative rods. The place from which the largest numbers of bacteria were found was the faucet of a sink in the corridor used by many students, and this was followed by students’ desks and the door handle of a bathroom entrance. A colony of S. aureus was found from one place. Although MRSA was not isolated from any of the places tested, four methicillin coagulase-negative Staphylococcus (MRCoNS) isolates were identified. This result indicates the environmental condition for the generation of MRSA through genetic exchange has already been constructed in the school. Based on the metagenomic method, S. aureus species were found in six different places, and MRCoNS was found in 15 different places. Interestingly, E. coli was identified in large numbers in all places except for one place, suggesting most of the places in the school had been contaminated by materials of fecal origin. Aside from the major pathogens, many of the opportunistic pathogens and fungi were isolated by the culture method. These consisted of the bacteria or fungi that can cause skin infection, endocarditis, urogenital infection, lung infection, allergy, and asthma in immunocompromised people and, for some species, even in healthy people. Thus, this study demonstrated that the high school students were continuously exposed to a risk of infection from the environment in which many of such bacteria and fungi were resident. In conclusion, this study is the first report that provides detailed information on the distribution of pathogens and opportunistic pathogens in the places students commonly frequent in the school. It is suggested that the results of the study will contribute to improving students’ healthcare in the school, providing the school with basic information on the common pathogens in the school environment, and thus promoting efforts for the more effective prevention of potential infections with major pathogens, including MRSA.
