LPS로 유도된 RAW264.7 세포에서 inducible nitric oxide synthase (iNOS)와 cyclooxygenase-2 (COX-2) 단백질의 발현과 tumor necrosis factor-α (TNF-α)와 interleukin-6 (IL-6)과 같은 염증성 사이토카인의 발현량은 농도 의존적으로 감소하였으며, heme oxygenase-1 (HO-1)의 발현은 농도 의존적으로 증가함을 확인하였다. 또한 Western blot 실험 결과, 농도와 시간이 지나감에 따라 HO-1의 발현을 확인할 수 있어 HO-1에 의한 항염증 효과를 확인할 수 있었다. 이러한 결과들을 종합해 볼 때, 파프리카 열매와 잎의 에틸아세테이트 분획물은 HO-1 발현을 통하여 RAW264.7 세포에서 항염증 및 세포보호 효과를 나타낸다는 것을 확인하였고 이러한 결과를 토대로 파프리카의 열매와 잎은 염증반응에 의한 만성질환을 예방할 수 있는 건강식품 소재로서 이용될 가치가 충분할 것으로 판단된다.

In order to investigate which extract or fraction and which part of paprika contains more functional compounds, antioxidant activities of 70% ethanol (EtOH) extract, hexane, chloroform (CHCl3), ethyl acetate (EtOAc), and water (DW) fractions of red, orange and yellow paprika fruits and leaf were analysed, respectively. And their anti-inflammatory effect were measured by LPS through MTT assay and NO assay. Anti-oxidant and anti-inflammatory effects were in the order from ethyl acetate> CHCl3> 70% EtOH> hexane> DW, and red> orange> yellow and the effects of leaf was as high as that of red. Since there is no cytotoxicity under 100 μg/mL, further anti-inflammatory experiments using RT-PCR and Western blot analysis were conducted using red paprika and leaf ethyl acetate fractions under 100 μg/mL. The red paprika and leaf ethyl acetate fractions suppressed the expression of proteins such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) and the expression of inflammatory mediators such as tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) and increased the expression of heme oxygenase-1 (HO-1) in a dose-dependent manner in a macrophage RAW264.7 stimulated by LPS using RT-PCR, According to the Western blot, the expression of HO-1 in a dose-dependent and time-dependent manner confirmed the anti-inflammatory effect of HO-1. From these results, it was confirmed that ethyl acetate fractions of red paprika fruit and leaf had anti-inflammatory and cytoprotective effect through the expression of HO-1 in a RAW264.7 cell and it was suggested that they might be used as health food materials to prevent the chronic diseases according the inflammatory response.
