Influenza A viruses (genus Influenzavirus A, family Orthomyxoviridae) exhibit a broad host spectrum and are able to infect birds, pigs, humans and several other mammals. The natural reservoir of influenza A viruses is wild waterfowl. Eighteen haemagglutinin (H) and eleven neuraminidase (N) subtypes have been identified so far. This study was determined to evaluation of pathogenicity and ability of transmission derived from animals, and development of insights of antiviral strategies need for control and prevention of influenza virus. The overall objectives of this thesis are: (a) prevalence and molecular characterization of animal influenza virus in wild bird, live bird market, poultry farm and pig; (b) to evaluate pathogenicity and understand characterization through mammalian host adaptaion of influenza H9N2 virus; (c) to examine the efficacy of polysaccharide of Persimmon Leaves (PLE0) against infection with mouse adapted A (H1N1) virus. The first, a combination of the following transmission mechanism may be a likely explanation for the spread of influenza virus by trade of live animals, the movement of animals within production systems, spread via feed, water, or equipment, migration of wild birds’s, people movement. However, the mechanisms driving this variation in animals have yet to be tested. To address these questions, we sampled a key influenza virus host species: domestic poultry farm, live bird markets, wild birds and pigs on a spatial scale, comprehensively throughout a complete annual cycle, measuring both current and past infections. We collected 20,483 samples and 274 samples yielded high pathogenic influenza virus and low pathogenic influenza virus. Of 274 influenza virus, various subtypes were co-circulation in LBM, wild bird, poultry and pig, major subtype was H9N2 virus. Our phylogenetic analysis of the HA gene segment of the Korean H9N2 viruses collected since 1996 and revealed the occurrence of different introductions within the country by poultry or wild birds. However, only a single lineage of virus evolved and circulated extensively in Korean poultry. The origin dates of the most recent common ancestors suggest that they emerged almost simultaneously probably from the MS96/96-like group. No evidence of new introductions of H9N2 strains from Asia to Korean poultry during 2001–2015 was detected, as shown by our neighbor-joining phylogenetic tree of the HA sequences representative of the H9 lineages. Enhanced surveillance of H9N2 viruses is needed to identify further increments in viral evolution and such data will help in a more timely update of vaccine strain to antigenically more closely match the circulating field viruses. These facts prompted us to assess the biologic properties and pandemic potential of H9N2 influenza viruses circulating in poultry. For the second objective, since their introduction during the late1996, these H9N2 viruses have evolved tremendously via intra- and inter subtype reassortment and genetic drift to become unique and distinct from the prototype MS96 viruses. Few studies have compared the biological properties of different H9N2 clades and determined whether certain clades pose a higher risk to mammals especially Korean lineage, and almost all of those have focused on G1 viruses isolated from China. To fill this gap in knowledge, we undertook an extensive characterization and risk-assessment study of H9N2 viruses on the pathogenesis, virulence and transmission of South Korea H9N2 viruses. We reported an H9N2 viruses isolated in South Korea, with higher than expected lethality in mice. These eight H9N2 viruses were various pathogenicity to mice which C21 and W25 were high pathogenic, C54 and SL-4P were middle pathogenic, W7 and C16 were low pathogenic, and C14 and C9 were non pathogenic. Especially, C21 and W25 showed highly lethal and could cause 90–100% mortality without prior adaptation after exposure to virus. Severe respiratory synderomes associated with diffuse lung injury, severe pneumonia and alveolar damage were observed in clinical affected mice. This finding emphasizes the necessity for careful, continual and thorough surveillance paired with risk-assessment of circulating influenza viruses. In the third aim, mice were oral administrated with polysaccharide of Persimmon Leaves (PLE0). Oral administration of PLE0 conferred a survival benefit to mice infected with the H1N1 influenza virus, and resulted in attenuation of viral replication in infected lungs. Influenza virus-infected mice receiving the PLE0 treatment show reduced mortality, which 100 mg PLE0 increased 20% survival rate after the influenza A virus infection. This result shows that the PLE0 is a potential candidate to fight influenza A virus infection. This extract may be a potent antiviral agent against influenza virus with no host toxicity. PLE0 is a potential therapeutic polysaccharide for the treatment of influenza virus infection and prophylaxis. Collectively, the finding of this studies exhibited that distribution and animal variation in prevalence of influenza A virus in South Korea and pathogenicity and risk fitness of H9N2 viruses would provide helpful to understand the risk of public health problem and important analysis such as virulence factor and evolution pattern. Furthermore, a potential therapeutic polysaccharide of Persimmon Leaves (PLE0) would provide helpful to prevent influenza A virus. Overall these studies offer an approach to control epidemic and edemic influenza A virus.
