본 연구에서는 뛰어난 효과를 나타내는 복합생균제를 제조하기 위하여, 고농도의 생균수와 대사물질이 풍부하게 함유된 발효방법 및 발효특성에 대하여 조사하였다. 복합생균제 제조에 적합한 B. subtilis B20, P. acidilactici L01, L. plantarum L30과 S. cerevisiae Y06를 각각 닭의 분변, 닭의 분변, 닭의 맹장과 돼지분변으로부터 분리 동정하였다. 분리된 미생물을 고농도 복합생균제로 이용하기 위하여 미생물의 접종방법을 달리하였으며, 그 결과 바실러스(B. subtilis B20)는 단독 발효, 유산균(P. acidilactici L01, L. plantarum L30)과 효모(S. cerevisiae Y06)는 혼합 발효하여 사용하였다. 고농도 생균수로 발효하기 위하여 원료, 첨가물 종류 및 농도, 온도, 수분함량에 대하여 조사하였다. 그 결과 대두박, 당밀 2%, 35℃, 수분 50%의 발효조건에서 발효 시, 고농도의 생균수가 함유되는 것으로 나타났으며, 바실러스와 유산균+효모 발효조건은 동일하게 나타났다. 복합생균제의 대량 생산을 위한 발효방법으로 회전식 드럼발효기, 트레이, 비닐 백, 쌀 포대를 이용하여 200kg 씩, 각각 바실러스와 유산균+효모를 발효하였으며, 그 결과 트레이 발효 시에 모든 생균수가 가장 높게 나타났다. 이와 같이 대량 발효 과정 시 생균수에 대한 검사 뿐 만 아니라, 항영양인자와 생리활성에 대한 조사를 하여 조단백질, 총 당, SDS-PAGE, pH, β-mannanase, 총 폴리페놀, isoflavone과 같이 생균수 이외에 점검해야 할 총 7개의 품질관리 지표를 설정하였다. 발효공정에 의하여 조단백질, 인, 칼슘, 조사포닌, 소화효소, 유기산, 항산화활성, 총 폴리페놀과 총 플라보노이드와 총 이소플라본배당체의 함량이 증가하였으며, 항영양인자의 함량은 감소하여 발효에 의하여 대사산물과 이용성이 증가되는 것으로 나타났다. 또한 발효공정 뿐만 아니라 저장조건과 저장기간에 대한 생리활성 변화를 조사하여, 저장기간 중에 생균수 이외에 점검해야 할 품질관리 지표로 0.2% KOH 용해도, pH, β-mannanase, 총 플리페놀 및 항산화 활성 중 ABTS radical 소거능을 포함한 총 5개의 항목을 설정하였다. 발효공정을 통해 제조된 복합생균제를 산란계에 급여하였을 때의 생산성, 계란품질, 맹장미생물, 장기 및 혈액에 미치는 영향, 혈액 내의 면역효과 및 계란저장성을 평가하여 생균제로 이용 가능성을 연구하였다. 그 결과 전체구간에서 난 생산성 지표인 산란율, 난중, 일산란양 및 사료효율은 대조구보다 높게 나타났으며(P<0.05), 전체구간에서 사료섭취량은 대조구보다 높은 경향을 나타내었다(P<0.10). 전체구간에서 계란품질 지표인 난각 두께, 난각 강도, 난황색 및 haugh unit은 처리 간 차이가 없었으며, 난각색은 0.5% 첨가구에서 높게 나타났다(P<0.05). 체내 조직의 간장, 비장, 심장, 사낭, 공장, 회장, 맹장, 복강지방, 난소관, 생체중의 중량에는 처리 간 차이가 없었으며, 공장의 길이는 첨가량이 증가할수록 감소하였다(P<0.05). 또한 장내 균총과 혈액 성분, 혈액 내 콜레스테롤에는 처리 간 차이가 없었다. 혈액 내 면역지표 성분인 IgA은 0.5% 첨가구, IgM은 0.3와 0.5% 첨가구에서 높게 나타났으나(P<0.05), IgG은 처리 간 차이가 없었다. 저장기간 내 Haugh unit은 4주차에 0.1% 첨가구에서 높은 경향을 나타내었으나(P<0.10), MDA는 처리 간 차이가 없었다. 본 연구의 결과는 최적화된 발효공정에 의해 제조된 산란계 사료 내 복합생균제의 급여가 생산성, 난각색 및 면역력을 증가시켰으며, 저장성을 향상시키는 경향을 나타내어 산란계의 생산성 개선에 도움을 줄 것으로 사료된다.

The present study investigated a fermentation process to produce probiotic strain containing high density of vegetable cell and metabolites in terms of development of high throughput probiotic mixture. Prior to investigation of method, isolation and identification of strain candidates were performed and B. subtilis B20, P. acidilactici L01, L. plantarum L30 and S. cerevisiae Y06 were isolated. To develop isolated strain for high cell density probiotic mixture, inoculation process was examined and it was detected that B. subtilis B20 was decided to be applied in single fermentation and lactic acid bacteria(P. acidilactici L01, L. plantarum L30) and yeast strain(S. cerevisiae Y06) were decided to be applied to co-fermentation. Effect of medium ingredients, different additives and their concentration, temperature and moisture contents were investigated to drive the fermentation process with high cell density. As a result, it was found that soybean meal, 2% of molasses, 35C and 50% of moisture were relevant to high cell density after fermentation in both of singular fermentation of B. subtilis and co-fermentation of lactic acid bacteria and yeast. For the scaling up of probiotic mixture, rotary and drum type, tray type and mash bag type fermenters were investigated. In results, tray type fermentation showed the greatest vegetable cell density. For the scaling up fermentation process, crude protein, total saccharides, protein gel electrophoresis(SDS-PAGE), pH, β-mannanase, total polyphenol and total isoflavone were measured for investigation of anti-nutrient factors and physiological activity and then those results were applied to quality control factors. Furthermore, activity alteration along with storage condition and duration were investigated. As a result, solubility in 0.2% KOH, pH, β-mannanase, total polyphenol and antioxidant activity(ABTS radical scavenging activity) were measured to additional check-factor for quality control to viable cell estimation. Manufactured probiotics, were then, fed to laying hens and its effect on productivity, egg quality, cecum and intestined microbes, and blood chemical parameter including immunoglobulin content and storage of egg were evaluated(p<0.05). Egg production, egg weight, egg mass and feed efficiency of treated group hens were higher than those of control hens(P<0.05). There was increasing tendency in feed in take when compared to control group(P<0.10). The egg shell color was significantly higher at 0.5% treatment than control group(P <0.05). The plasma IgM in laying hens fed the probiotics supplemented at the lead of dietary 0.3% of 0.5% was higher than that of control(P<0.05). Dietary 0.5% probiotics showed higher IgA concentration than control hens(P<0.05). The haugh unit of hens fed the mixed probiotics was tended to be increased when compared with the control group, after 4 weeks of storage(P<0.10). The level of malondialdehyde(MDA) were not significantly different among the groups. All together, feeding probiotics showed improvement in productivity, egg shell color and immunity and prolonged storage of egg by feeding probiotics. There was no negative influence on physiological status of laying hen and it was suggested that the probiotics can give a beneficial effect on productivity of laying hen as result shown above.
