Saposhnikovia divaricata, belonging to Umbelliferae family, is mainly distributed in China and Japan. The roots of S. divaricata have been used traditionally as analgesics, antipyretics and antiphlogistics. Phytochemical studies have revealed that this plant contains coumarin, chromone and polyacetylene. The dried roots of S. divaricata were extracted with MeOH, and partitioned with n-hexane, CH2Cl2, EtOAc, and water, successively. The n-hexane and CH2Cl2 soluble fractions ware purified by several column chromatography including MPLC and preparative HPLC. The structures of the isolated compounds were elucidated as umbelliferone (1), scopoletin (2), psoralen (3), xanthotoxin (4), bergapten (5), isopimpinellin (6), phellopterin (7), imperatorin (8), isoimperatorin (9) marmesin (10), deltoin (11), anomalin (12), (E)-5-methoxytrichoclin (13), (E)-5-methoxytrichoclin acetate (14), cleomiscosin A (15), noreugenin (16), hamaudol (17), 3′-O-angeloylhamaudol (18), 3′-O-acetylhamaudol (19), 3′-O--D-glucosylhamaudol (20), melanochromone (21), ledebouriellol (22), divaricatol (23), 1″-O-glucosylcimifugin (24), norcimifugin (25), visaminol (26), 5-O-methylvisaminol (27), 4′-O--D-glucosyl-5-O-methylvisaminol (28), monnieriside G (29), falcarindiol (30), panaxynol (31), atractylenolide II (32), atractylenolide III (33), tyrosol (34), vanillin (35), 2-methoxy-2-(4′-hydroxyphenyl)ethanol (36), and -hydroxypropiovanillone (37) by comparing their physicochemical and spectroscopic data with those of published values. Among them, compounds 12, 22, 30, and 31 showed inhibitory activity against LPS-induced NO production in RAW264.7 cells with their IC50 values ranging from 6.5 μM to 17.1 μM. These results suggested that coumarin, chromone and polyacetylene, from S. divaricata are worthy of further investigation for their potential as anti-inflammatory agent.
