The ubiquitin-proteasome system (UPS) and the autophagy-lysosome system are two major protein degradation systems that crosstalk and coordinate their roles in the removal of misfolded proteins. Proteasomal inhibition leads to the induction of autophagy, which is mediated by multiple biological pathways. Prior to this investigation, the involvement of the N-end rule pathway in this crosstalk has never been reported. Our recent study has shown that the N-end rule pathway mediates selective autophagy of misfolded proteins in a ubiquitin-independent mechanism under stimulation with cytoplasmic double-stranded DNA (dsDNA). In this mechanism, cytoplasmic dsDNA induces N-terminal arginylation of BiP/GRP78/HSPA5, an ER chaperone, in the cytoplasm where it binds misfolded proteins and p62. The binding of p62 ZZ domain with N-terminal arginine of BiP facilitates p62 polymerization which leads to the targeting of p62-R-BiP-misfolded protein complexes to autophagosomes. After fusion with lysosomes, the complexes undergo lysosomal degradation. Here, this study show that like cytoplasmic dsDNA, stimulation with proteasome inhibitors induces N-terminal arginylation of BiP (R-BiP). R-BiP forms cytoplasmic puncta that co-localize with p62 and LC3, positive autophagosomes, indicating p62–mediated delivery of R-BiP to autophagosomes. Inhibition of autophagic flux revealed that proteasome inhibition-induced R-BiP is degraded by autophagy. Upon proteasome inhibition, R-BiP puncta also co-localize with ubiquitin conjugate puncta, and their formation and autophagic degradation are dependent on p62, suggesting the role of R-BiP and p62 in protein quality control. Interestingly, adequate induction of autophagosomes upon proteasome inhibition is also dependent on p62. In fact, knocking down or knocking out p62 significantly diminished the level of LC3-II in both control cells and proteasome inhibited cells. Using autophagic flux assays, these data shown that the lower level of LC3-II in p62 knockout mouse embryonic fibroblasts (MEFs) is due to a reduction in the basal level of LC3. This study has identified p62 as a key mediator of crosstalk between the ubiquitin-proteasome system and autophagy.
