Accordingly, we prepared four recombinant influenza A viruses that contain a single amino acid mutation by reverse genetics system and compared their virulence upon mouse infection. It was observed that the virus with a valine-to-alanine mutation at position 180 of NS1 caused mouse lethality most dramatically among them (67% dead). It is noteworthy that even though the recombinant virus with a mutation of alanine to glutamic acid at amino acid 377 in PB2 showed no effect on lethality, combination of these two mutations further decreased the mouse survival rate (100% dead). Given that these two mutations co-exist in the viral isolate HK-P15B, it can be suggested that they play a role for causing mouse virulence synergistically by reducing innate immune response by the mutant NS1 as well as by upregulating viral RNA replication efficiency by the mutant PB2. Moreover, the relationship between of mouse lethality and these two amino acid mutations were confirmed again by histological analysis and molecular biological studies using influenza A virus-infected mouse lung tissues. The present study suggests that the mutations of V180A in NS1 and A377E in PB2 are major determinants for virulence in mammals. This finding could be applied to prediction of pathogenicity of an emerging or re-emerging influenza A viruses and to defense against influenza pandemics. In addition, the mouse-adapted influenza viral strain, HK-P15B, could be useful for evaluation of efficacy of antiviral or vaccine candidates in vivo.
