Bacteriophages have been proposed as new tools to control pathogenic bacteria. In this study, two Listeria bacteriophages, LMP1 and LMP7, were isolated from chicken feces as biocontrol of L. monocytogenes. The host specificity of these two bacteriophages were determined by double-layer methods with 5 different serotypes of L. monocytogenes strains. Both bacteriophages had a lytic effect on L. monocytogenes ATCC 7644, 15313, 19114, and 19115. The Lytic activity of Listeria bacteriophages was assessed in TSB contaminated with 5×105 CFU/ml L. monocytogenes, ATCC 7644 or ATCC 19114, and incubated at 30℃ for 24 hours. The Phage lytic activity was also evaluated at 10℃ for 5 days in the same composition. LMP1 and LMP7 were able to inhibit the growth of L. monocytogenes ATCC 7644 and 19114 at 30℃ and 10℃ condition. Nevertheless, LMP1 was more effective than LMP7 in hosting L. monocytogenes ATCC 19114. On the contrary to this observation, LMP7 was more effective than LMP1 in hosting L. monocytogenes ATCC 7644. Therefore, LMP1 has a low lytic activity for L. monocytogenes ATCC 7644. The morphology of LMP1 and LMP7 showed resemblance to the members of the Siphoviridae family. The bacteriophages genome sequence was determined by next-generation-sequencing (NGS) methods. These two phages had a double-stranded circular type DNA. The LMP1 genome contains 40,867 bp and 69 coding sequence (CDS) gene and 36.84% G+C content. The LMP7 genome contains 41.714 bp and 67 CDS gene and 36.8% G+C content. The ability of LMP1 and LMP7 to inhibit the growth of L. monocytogenes ATCC 7644 and 19114 was also evaluated in milk. In this experiment, milk was inoculated with 5×105 CFU/ml of L. monocytogenes, ATCC 7644 or ATCC 19114, and was incubated at 30℃ for 20 hours and at 10℃ for 5 days. The growth of L. monocytogenes ATCC 7644 was inhibited by both LMP1 and LMP7 in milk. In contrast, the growth of L. monocytogenes ATCC 19114 was only inhibited by LMP1. The bacteriophages lytic activity was also evaluated at 4℃ in milk in order to confirm the potential use of these phage at refrigerated food. In conclusion, the two Listeria bacteriophages, LMP1 and LMP7, were isolated. These two bacteriophages exhibit different host specificities and characteristics, thus they can be used effectively as a component of phage cocktail to control L. monocytogenes in the food industry.
