CD44는 자신과 결합하고 있는 c-Src 등을 통해 하류반응을 조절함으로써 발암과 관련된 신호전달을 매개한다. 유방암세포에서 CD44의 기능을 알아보기 위해, CD44에 대한 레트로바이러스 매개의 shRNA를 이용하여 CD44의 발현이 억제된 세포주를 만들었다. CD44의 발현 억제를 통해 유방암세포의 성장, 이동, 침윤 등이 감소하는 것을 확인할 수 있었고, 세포 이동과 관련된 단백질인 c-Src, paxillin, FAK의 발현 및 활성 또한 감소하는 것을 확인할 수 있었다. 또한 CD44의 발현이 억제된 세포주에서 GSK3β 의존적인 단백질 분해 경로의 촉진을 통해 c-Jun 단백질의 발현이 음성적으로 조절된다는 것을 확인하였고 이로 인해 c-Jun의 표적 단백질인 Sp1의 발현이 억제된다는 것을 관찰하였다. 마지막으로, CD44 발현 억제 세포주에서 볼 수 있었던 c-Src의 전사인자인 Sp1의 발현 감소는 c-Src의 단백질뿐만 아니라 mRNA의 발현 억제를 유발함으로써 MAPK 신호 전달 경로의 감소를 초래하였다. 최종적으로, 인간 유방암세포에서 CD44 발현 억제로 인해 관찰할 수 있었던 생물학적 변화들은 c-Jun, Sp1, c-Src의 순차적인 하향 조절에 의해 매개되었다는 것을 의미한다. CD44와 관련된 많은 연구들은 암줄기세포 바이오 마커로써의 CD44 기능에 더 초점을 맞춰왔다. 그렇기 때문에 CD44가 암세포 대사작용에 어떤 영향을 주는지에 대한 연구는 아직 미흡하다. 따라서 본 연구자는 CD44에 의한 암세포 대사조절 기능을 알아보기 위해 CD44의 발현이 억제된 세포주를 만들었고, 해당 표현형을 확인한 결과, 포도당 흡수능력, ATP 생성능력, 젖산 생성능력이 모두 감소하는 것을 볼 수 있었다. 또한 CD44 발현 억제로 인한 젖산탈수소효소(lactate dehydrogenase, LDH)의 발현이 LDHA 발현은 감소하고 LDHB의 발현은 증가함에 따라 LDH1 쪽으로 이동하는 것을 볼 수 있었다. 이는 곧 젖산탈수소효소의 발현 패턴이 암세포 대사조절에 중요한 영향을 미친다는 것을 의미한다. These cells do not respond well to ER-targetted hormone therapy or medications that block HER2. Therefore, TNBC patients show poor prognosis and tend to exhibit lower survival rates compared to other subtypes of breast cancer. The purpose of this study was to find therapeutic target for TNBC. CD44 which has been well known as breast cancer stem cell marker and known to be overexpressed in TNBC cell lines, MDA-MB-231 and Hs578T. The cytoplasmic tail of CD44 is known to bind ERM (ezrin, radixin, moesin) proteins, cytoskeletal proteins like ankyrin, and the non-receptor tyrosine kinase c-Src. CD44 transmits its oncogenic signaling via c-Src and its downstream effectors. To investigate the role of CD44 in breast cancer cells, I generated CD44 knock-down cells via retroviral delivery of shRNA against CD44. I found that silencing of CD44 decreased the proliferation, migration, and invasion of breast cancer cells. The expression and activity of cell migration-related proteins, including c-Src, paxillin, and FAK was decreased by CD44 silencing. I also found that the c-Jun protein level was negatively regulated via induction of a GSK-3β-dependent degradation pathway in CD44 knock-down cells. The expression level of Sp1, a target gene product of c-Jun, was also decreased in these cells. Finally, CD44 knock-down suppressed both mRNA and protein levels of c-Src and its downstream MAPK pathway as a result of down-regulation of Sp1 as a transcription factor for c-Src. These results indicate that biological changes induced by CD44 silencing are mediated by cumulative down-regulation of c-Jun, Sp1, and c-Src in human breast cancer cells. Although many studies have focused on CD44 as a cancer stem cell marker, its effect on cancer cell metabolism remains unclear. The Warburg effect is specialized metabolism of cancer cells, which allow cancer cells to generate ATP more rapidly than oxidative phosphorylation. I found that silencing of CD44 decreased the glycolytic phenotype of cancer cells, affecting glucose uptake, ATP production, and lactate production. I also found that ablation of the CD44-induced LDH isoenzyme results in a shift to LDH1 due to LDHA down-regulation and LDHB up-regulation, implying the importance of LDH isoenzyme modulation on cancer metabolism. The expression of glycolysis-related proteins including HIF-1α and LDHA was decreased by CD44 silencing. These effects were due to the up-regulation of LKB1/AMPKα activity by reduction of c-Src and Akt activity in CD44 knock-down cells. Finally, induction of LKB1/AMPKα activity blocked the expression of HIF-1α and its target gene, LDHA. Inversely, LDHB expression was repressed by HIF-1α. These findings indicate that the CD44 silencing-induced metabolic shift is mediated by regulation of c-Src/Akt/LKB1/AMPKα/HIF-1α signaling in human breast cancer cells. Interestingly, I found that galectin-1 (Gal-1) was down-regulated in CD44 knock-down cells. Galectin-1 is a β-galactoside binding protein secreted by many types of aggressive cancer cells. Although, there have been many researches performed on galectin-1 functions in cancer progression, relatively little attention has been focused to galectin-1 as a cell surface therapeutic target. To elucidate the molecular mechanisms underlying galectin-1-mediated cancer progression, I established galectin-1 knock-down cells via retroviral delivery of shRNA against galectin-1 in triple-negative breast cancer (TNBC) cell lines, MDA-MB-231 and Hs578T. Ablation of galectin-1 decreased cell proliferation, migration, invasion, and doxorubicin resistance. I found that such phenomena were caused by a decrease in galectin-1-integrin β1 interaction and suppression of the downstream FAK/c-Src pathway by galectin-1 ablation. I also found that silencing of galectin-1 inhibited ERK/STAT3 signaling which results in blocking of survivin expressions, implying the STAT3 as a transcription factor for survivin. Finally, rescue of the endogenous galectin-1 knock-down and recombinant galectin-1 treatment recovered FAK/c-Src/ERK/STAT3/survivin pathway. Taken together, the results suggest that cell surface galectin-1 contributes to cancer progression and doxorubicin resistance in triple-negative breast cancer cells, which might be ascribed to galectin-1-induced up-regulation of integrin β1/FAK/c-Src/ERK/STAT3/survivin pathway. My results imply that cell surface galectin-1 may act as a potential therapeutic target for triple-negative breast cancer.

To elucidate the molecular mechanisms underlying galectin-1-mediated cancer progression, I established galectin-1 knock-down cells via retroviral delivery of shRNA against galectin-1 in triple-negative breast cancer (TNBC) cell lines, MDA-MB-231 and Hs578T. Ablation of galectin-1 decreased cell proliferation, migration, invasion, and doxorubicin resistance. I found that such phenomena were caused by a decrease in galectin-1-integrin β1 interaction and suppression of the downstream FAK/c-Src pathway by galectin-1 ablation. I also found that silencing of galectin-1 inhibited ERK/STAT3 signaling which results in blocking of survivin expressions, implying the STAT3 as a transcription factor for survivin. Finally, rescue of the endogenous galectin-1 knock-down and recombinant galectin-1 treatment recovered FAK/c-Src/ERK/STAT3/survivin pathway. Taken together, the results suggest that cell surface galectin-1 contributes to cancer progression and doxorubicin resistance in triple-negative breast cancer cells, which might be ascribed to galectin-1-induced up-regulation of integrin β1/FAK/c-Src/ERK/STAT3/survivin pathway. My results imply that cell surface galectin-1 may act as a potential therapeutic target for triple-negative breast cancer.
