DPPH 라디컬소거 활성은 0.2 mM 2,2-diphenyl-1-picry-lhygrazyl (DPPH) 1 ㎖에 High-purity water, 수돗물, 플라즈마 방전수, ascorbic acid를 200 ㎕ ~ 800 ㎕씩을 혼합하여 23℃에서 10 min 동안 반응 시킨 후 ELISA reader 사용하여 517 nm에서 흡광도를 측정하였다. 플라즈마 생성장치의 최종 양산 제품이 나온 후 초기 세균수 1 x 105~106 CFU/mL인 E. coli 만 플라즈마 방전수에 30±5s 노출시켜 희석평판법으로 사멸율을 측정하여 플라즈마 방전수의 효능을 평가 하였다. 양산 제품 결과 역시 방전수 처리후에 생균의 숫자가 <10 CFU/ml 나타나 99.9999% 살균력을 보여서 플라즈마 생성장치가 E. coli 등 4개의 세균에 우수한 대한 살균력 효능이 있음을 보여주었다.

The present study investigated the bactericidal effect of the plasma water discharged from tap water using small electrical discharge apparatus consisted of the cell layer supplied with 5V. This plasma water contains various types of ROS that can be detrimental on bacteria. In this study, the experimental bacterial strains included Escherichia coli, Pseudomonas aeruginosa, Salmonella enteritidis, Staphylococcus aureus, Streptococcus mutans. These bacteria were exposed to the plasma discharged water for 30±5s followed by dilution plate method. The initial concentrations of bacteria were approximately adjusted to be 1 x 105∼106 CFU/ml. The population of all bacteria tested decreased down to less than 10 CFU/ml showing 99.999% ∼ 99.9999% death rates except for S. mutans. The bactericidal activity of S. mutans was less compared to the other strains, showing reduction from initial concentration of 1 x 105∼106 CFU/ml to 185 CFU/ml indicating 99.98∼99.998% death rate. In addition, DPPH assay as a simple and rapid method was conducted to measure indirectly ROS of plasma discharged water. Test of antibacterial activity using dilution plate is unrealistic in industrial situation because bioassay using dilution plate method requires microbial expertise, equipments, and laborious works and time consuming. Removal activity of radicals was conducted using 0.2 mM DPPH. The one ml of 0.2 mM DPPH was mixed with plasma discharged water, highly purified water, tap water and ascorbic acid (Vit C) with ratio of 1:0.2, 1:0.4, 1:0.6, and 1:0.8 for each sample, respectively. The reaction mixtures were incubated for 10 min at 23℃ and the absorbances at 517 nm were measured. The absorbances of plasma discharged water were significantly different from tap water depending on the ratio of DPPH and sample mixture, showing 0.757 and 0.470 of plasma discharged water and tap water, respectively, suggesting that this method could be applied to quality test for plasma discharged water apparatus in industrial situation.
