In this study, it was carried out to investigate the effects of Stachys sieboldii Miq root powder on anti-obesity and cholesterol lowering effects in rats fed a high fat-high cholesterol diet for 6 weeks. It was also conducted to investigate major nutrients component and bioactive materials of Stachys sieboldii Miq root powder. The proximate compositions of Stachys sieboldii Miq root as a dry matter basis were 4.27% of moisture content, 15.04% of crude protein, 0.98% of crude fat, 4.62% of crude ash and 75.09% of carbohydrate, respectively. Analysing free sugar, major free sugars were identified as sucrose, glucose and fructose. Total amino acid content of Stachys sieboldii Miq root was 11,079,831 mg%, and essential amino acid was 3,261.089mg%. Major amino acids of Stachys sieboldii Miq root were glutamic acid, aspartic acid, threonine, arginine and lysine. Analysing total fatty acids, 26 kinds of components were isolated from Stachys sieboldii Miq root. Major fatty acids were linoleic acid, palmitic acid and linolenic acid. Malic acid, citric acid and formic acid were the major organic acids. As a result of vitamin analysis, the contents of vitamin C was the highest among the vitamin in Stachys sieboldii Miq root. The major components of mineral were K, Ca, Mg. Total polyphenol and flavonoid content of Stachys sieboldii Miq root ethanol extract were found 20.44 mg/g, 11.510 mg/g in 1,000 ppm. The anti-obesity and cholesterol lowering effects of Stachys sieboldii Miq root powder in rats fed a high fat-high cholesterol diet. Experimental groups were normal diet group(N), high fat-high cholesterol diet group(HFC), high fat-high cholesterol diet with 3% Stachys sieboldii Miq root powder group(HFC-SSL) and high fat-high cholesterol diet with 5% Stachys sieboldii Miq root powder group (HFC-SSH). The body weight gain and FER were increased due to a high fat-high cholesterol diet, but gradually decreased in the Stachys sieboldii Miq root powder fed groups compared with the HFC group. The liver and eididymal, mesenteric, retroperitoneal, perinenal and total adipose tissues weight of HFC group were heavier than that of N group, whereas those of groups administered Stachys sieboldii Miq root powder were gradually decreased. The serum ALT, AST, ALP and LDH activitie were signicantly decreased in the Stachys sieboldii Miq root powder fed groups compared with the HFC group. Levels of serum triglyceride(TG), total cholesterol(TC), LDL-C, atherogenic index and cardiac risk factor decreased in the Stachys sieboldii Miq root powder fed groups compared with HFC group, while the serum HDL-C level decreased in the HFC, but this level increased significantly in the Stachys sieboldii Miq root powder fed groups. Levels of total cholesterol and triglyceride in liver and adipose tissues were lower in Stachys sieboldii Miq root powder administered groups than HFC group. High fat-high cholesterol diet administration were induced the marked accumulation of large lipid droplets and resulted in typical histopathology, but Stachys sieboldii Miq root powder administered groups attenuated lipid droplets accumulation in hepatocytes. Fat cell size of epididymal adipose tissue was lower in Stachys sieboldii Miq root powder administered groups compared with the HFC group. The mRNA expression in river that are considered to play an important role in lipid and cholesterol metabolism effect. ACC, FAS, G6PDH, HMG-CoA reductase mRNA expression of Stachys sieboldii Miq root powder administered groups in river were significantly decreased compared to those of HFC group. CYP7A1, LDL-receptor mRNA expression was one the highest in the high fat-high cholesterol diet with 5% Stachys sieboldii Miq root powder group (HFC-SSH). The results suggest that Stachys sieboldii Miq root powder could make better lipid metabolism of serum, liver and adipose tissue and may decrease lipid storage. Therefore Stachys sieboldii Miq root powder may inhibit fatty tissue of the body and lower blood cholesterol levels shown preventive effect of improving lipid metabolism and metabolic diseases prevention.
