The glutamate decarboxylase (GAD) catalyzes the production of GABA by removing carbon dioxide from L-glutamate or monosodium glutamate. To date, GADs have been reported mainly from Lactobacillus brevis species. In the present study, two novel genes encoding GADs (BfdGAD and LbaGAD) were cloned from Bifidobacterium dentium KCTC 3362 (ATCC27679) and Lactobacillus antri KCTC 5855 (DSM16041), respectively. BfdGAD and LbaGAD consists of 476 and 468 amino acids, which share about 45% and 67% with those from Lactobacillus brevis, respectively. These recombinant GADs with C-terminal six histidines were purified by Ni-NTA column chromatography, and their enzymatic properties were comparatively characterized. BfdGAD showed the highest activity at 30oC and pH 4.0, while LbaGAD exhibited its maximal activity ac 60oC and pH 4.5. In the presence of 0.2 mM pyridoxal 5’-phosphate as an essential cofactor, the specific activity of BfdGAD and LbaGAD are 30.1 U/mg and 1.31 U/mg against 50 mM monosodium glutamate substrate, respectively. These GADs will be utilized for the enzymatic production of GABA in the food industry, as they are originated from the probiotic microorganism candidates.
