B-134 균주는 그람양성, 간균으로 MRS 고체배지상에서 연한 노란색을 띄며 circuler (appearance), entire (margin) 그리고 convex (elevation)인 집락의 특징을 나타내었다. 16S rRNA 염기서열을 이용한 계통분석에서 B-134 균주는 Lactobacillus plantarum subsp. plantarum ATCC 14817와 99.93%의 유사도를 나타내었다. 또한 B-134균주의 gadB 유전자의 염기 및 아미노산 서열분석을 통하여 Lactobacillus plantarum subsp. plantarum ATCC 14817 균주와 100%일치하는 것을 확인하였다. 결과적으로 B-134 균주는 형태, 기질이용능력, 16S rRNA 염기서열 및 gadB 유전자 분석의 결과로부터 Lactobacillus plantarum subsp. plantarum B-134 균주로 명명하였다. B-134 균주의 생장을 위한 최적조건은 온도 37℃, pH 6.1~6.7, 2% NaCl, 0~4% MSG로 나타났다. 반면 GABA 생성을 위한 최적배양조건은 온도 37℃, pH 5.7, 0% (w/v) NaCl, 3% (w/v) MSG로 조사되어졌다. 특히, GABA 생성을 위한 중요한 환경인자는 pH로 확인되었다. GABA 생산을 위한 최적 배양조건에서 GABA 농도는 증가되었고 배양 36시간째에 56 mM 까지 도달하였다.

Makgeolli-derived LABs were isolated 64 strains according to the characteristics of the shape and color of the colony grown on MRS agar plate. The GABA production ability of the isolated strain were cultured in MRS broth contained 1% MSG and evaluated by TLC and RP-HPLC analysis. As a result, B-134 strain was selected as a superior strain for GABA production. Strain B-134 was Gram positive, rod shaped and formed yellow, circuler (appearance), entire (margin), and convex (elevation) colony within 1 day on MRS agar plate at 30℃. Phylogenetic analysis based on 16S rRNA gene sequences indicated that this strain is related to members of the genus Lactobacillus and showed high sequence similarity to L. plantarum subsp. plantarum ATCC 14817 (99.93%). In the analysis of nucleotide and amino acid sequence of glutamate decarboxylase B (gadB), B-134 strain was confirmed to be 100% match with Lactobacillus plantarum subsp. plantarum ATCC 14817. From these results, strain B-134 was tentatively identified as a Lactobacillus plantarum subsp. plantarum B-134. The optimal condition of strain B-134 for cell growth was 37℃, pH 6.1~6.7, 2% NaCl, 0~4% MSG. Whereas, optimal condition for GABA production was 37℃, pH 5.7, 0% NaCl, 3% (w/v) MSG.
