The efficiency of transformation from the petiole and the leaf was approximately 12% and 27%, respectively. A total of 36 lines were confirmed by PCR amplification of rol genes. Of these, eight HR lines were selected for further study. Line TP4 derived from the petiole displayed the highest growth index (37.8). Among hairy root cultures derived from petioles, line TP2 accumulated the highest concentration of triterpenoid saponins (46.57 mg∙g-1). Triterpenoid saponins concentration was >2-fold higher in the leaves than in the petiole of the ex vitro plants. In addition, triterpenoid saponins content in HR cultures derived from the petiole was 1.4-fold higher than that derived from the leaf. Taken together, the results of this study confirmed that the triterpenoid saponins content of the origin tissue does not affect to triterpenoid saponins content of hairy root derived from genesis tissue.
