포스포라이페이즈 D(PLD)는 포스파티딜콜린을 가수분해하여서 포스파티딕 에시드(PA)를 만드는 역할을 하는 효소이다. Phospholipase D (PLD) is an enzyme that hydrolyzes phospholipids to yield phosphatidic acid (PA). PLD and PA play an important role in a various physiological processes and diseases. Specifically in cancer, PLD and PA have been implicated in cell proliferation, cell survival, migration, and invasion. PLD is considered as attractive therapeutic target for cancer because of its multiple implications in tumorigenesis. However no drugs, that target PLD, have been approved by the FDA until now. Recently, several studies have revealed that halopemide derivatives including FIPI and VU0155069 show inhibitory activity against PLD. Thus, fragment based drug design was performed utilizing the structures of FIPI and VU0155069 as a starting point, resulting in the development of B728 as a hit compound. Through the structural modification of B728, a total of 113 compounds were synthesized and biologically evaluated for their inhibitory activity against PLD in a cell based assay. More than 20 derivatives were identified as active compounds. Among them, compound 5j exhibited the most potent inhibitory activity against PLD than any other compounds. Subsequent studies including a safety study and an in vivo mouse model study were performed to verify toxicity and potency. Results from these studies indicated that compound 5j could not only be a promising PLD inhibitor for cancer therapy but also be used as a tool for elucidating the physiological role of PLD Acute myeloid leukemia (AML) is a clonal disorder of hematopoietic progenitor cells characterized by the accumulation of nonfunctional and immature blood cells (i.e., myeloblasts). In AML, a mutation in FLT3 is commonly occurs and is associated with poor prognosis. We have previously reported that thieno[2,3-d]pyrimidine derivative compound 1 exhibited better antiproliferative activity against MV4-11 cell line which harbors mutant FLT3 cells than AC220, which is a well-known FLT3 inhibitor, and has good microsomal stability. However, compound 1 had poor solubility. Then, further structural modification were carried out at the C2 and the C6 positions of thieno[2,3-d]pyrimidine scaffold. Compound 13b, which possesses a thiazole moiety at the C2 position, exhibited better antiproliferative activity than compound 1 and showed increased solubility and moderate microsomal stability. These results indicate that compound 13b could be a promising potential FLT inhibitor for AML chemotherapy and a precursor for further modification.
