본 연구에서는 전통발효식품에서 분리한 미생물, 미생물유래 효소를 이용하여 진세노사이드 Rb1 및 홍삼농축액에 포함되어있는 사포닌을 가수분해하고 기능성 사포닌 ginsenosides Rd, Rg3, Rh2 및 compound-K를 생산하고자 하였다. 20종의 전통발효식품을 수집하여 MRS media를 이용하여 약 200여종의 미생물을 분리하였다. 이중 가수분해 활성능이 있는 미생물을 구분하기 위하여 esculin agar를 이용하여 65개의 β-glucosidase활성 균주를 분리하였다. 각 분리한 균을 24~72h 액체 배양한 후 ginsenoside Rb1, 홍삼농축액과 1:1 비율로 각각 반응시켜 minor saponin, 즉 2차 대사산물로의 전환을 확인하였다. 분리된 몇몇 균주들은 Rd, Rg3, F2등의 생산이 가능한 활성을 나타내었으며, 그 효능 및 효과를 증대시키기 위하여 미생물 유래 효소인 multipact FE를 함께 처리 하였을 때, F2의 함량증가와 compound-K가 추가 생성되는 것을 확인할 수 있었다. Rd, Rg3, F2등으로의 전환 활성을 나타내는 균주 중에서 활성이 우수한 strain WJ-7을 동정한 결과 Lactobacillus paracasei subsp. paracasei.로 확인되었다. 본 연구 결과, 발효식품으로부터 분리된 Lactobacillus paracasei subsp. paracasei WJ-7은 진세노사이드 전환 활성을 가지고 있어 이를 적용한 건강기능식품으로서의 개발 가능성을 확인할 수 있었다.

The aim of this study was to isolate lactic acid bacteria from traditional fermented foods which can produces functional saponins, ginsenoside Rd, Rg3, Rh2 and compound K by hydrolyzing ginsenoside Rb1 and saponins of red ginseng concentrates. Several lactic acid bacterial strains were isolated from fermented food samples. Among the total 200 isolated strains, 65 strains showed β-glucosidase activity which was examined using esculin agar plates. The β-glucosidase producing strains were further incubated with the protopanaxadiol major saponin ginsenoside Rb1. As a result, some of the strains were showed the ability to hydrolyze the β-glucosyl linkages in ginsenoside Rb1. Amongst a strain designated as WJ-7 was exhibited the most potent ability to convert ginsenoside Rb1 into pharmacologically highly efficient minor ginsenoside Rg3. Moreover, the WJ-7 along with MPFE enzyme and red ginseng extract successfully synthesized into another pharmacologically active minor ginsenoside CK which conformed by TLC and HPLC. The similarity analysis based on 16S rRNA gene sequence revealed that the WJ-7 strain phylogenetically related to the genus Lactobacillus and it is closely related to Lactobacillus paracasei subsp. Paracasei. The growth characteristics of strain Lactobacillus paracasei subsp. paracasei showed that the highest growth was observed in 100% MRS medium with pH 6.5 to 7.0 for 24 hrs at 37 °C incubation temperature. In conclusion, in this study lactic acid bacteria isolated from fermented foods had ginsenoside conversion activity, and these might be utilized further to develop as functional food for betterment of healthy human kind.
