In this study, we examined the effects of PS on the stimulation of osteoblast differentiation and inhibition of osteoclast generation and its mechanism. The calvarial cells isolated from calvarie of mice were differentiated into osteoblast by ascorbic acid (AA) and β-Glycerophosphate (β-GP) and the effect of PS on stimulation of osteoblast differentiation was analyzed by alkaline phosphatase (ALP) and alizarin red s staining (ARS) assay. Bone marrow-derived macrophages (BMMs) were cultured with macrophage colony-stimulating factor (M-CSF) and receptor activator of nuclear factor kappa-B ligand (RANKL) to differentiate into osteoclast and the effect of PS on the inhibition of osteoclast generation was analyzed by tartrate-resistant acid phosphatase (TRAP) assay. Factors related with differentiation of osteoblast and osteoclast were identified by quantitative real-time PCR and western blot analysis. In conclusion, PS stimulated osteoblast differentiation and inhibited osteoclast generation without toxicity. The stimulatory effect resulted from the increased expression of ALP, bone sialoprotein (BSP), osteocalcin (OCN), osteopontin (OPN), and osterix, and the increased phosphorylation of Smad1/5/8, glycogen synthase kinase 3 beta (GSK3β), Akt, p38, extracellular signal-regulated kinase (ERK) and c-Jun N-terminal kinase (JNK). Also, PS increased the expression of Runt-related transcription factor 2 (Runx2) which is a key regulator of osteoblast differentiation. The increase in mineralization of osteoblast by PS was confirmed by ARS staining. The inhibitory effect of PS on osteoclast generation resulted from the decreased expression of osteoclast-associated receptor (OSCAR), TRAP, cathepsin K, ATPase, H+ transporting, lysosomal 38 kDa, and V0 subunit d2 (Atp6v0d2) via inhibition of c-Fos and nuclear factor of activated T-cell (NFATc1). Also, PS prevented RANKL mediated osteoclast differentiation by inhibiting phosphorylation of p38. Consequently, it is suggested that PS may be potential medication for bone diseases such as osteoporosis.
