본 연구는 혼합 목재분말의 열수 추출물을 활용하여 금과 은 나노입자의 생산방법 최적화, 특성분석, 그리고 이들 입자의 활용을 위하여 항균활성을 조사하였다. 혼합 목재분말의 조성을 확인한 결과 셀룰로오스와 헤미셀룰로오스를 구성하는 다양한 당성분이 존재하였으며, 독성 중금속의 함량은 존재하지 않거나 매우 미량으로 존재하였다. 두 종류의 금속 나노입자 합성은 표면 플라즈몬 공명 곡선 측정에 기반하여 측정하였으며, 연구결과 혼합 목재분말 추출물은 나노입자 합성에 환원제와 안정제 역할을 하였다. 은 나노입자의 경우 AgNO3와 목재추출물의 농도 의존적 합성율을 보였으며, pH가 11이 넘는 알칼리성 환경에서 매우 높은 합성율을 보였다. 그리고 최대 흡광도를 나타내는 파장은 pH가 증가함에 따라 감소하였으며, 이는 pH가 감소함에 따라 은 나노입자의 크기가 증가하거나 서로 응집함을 의미한다. 가시광선의 최적 광량과 최적 온도는 각각 100 μmole·m-2s-1과 30℃로 확인되었다. 금 나노입자 합성의 경우 은 나노입자 합성과 달리 최적 pH는 약산성 및 중성에 해당하는 pH 5-7 이었으며, HAuCl4의 농도가 1 mM 이상 증가할 경우 단분자 형태의 금 나노입자 형성이 진행되지 않았고 목재 추출물의 농도는 합성에 영향을 주지 못했다. 그리고 반응온도와 pH가 증가함에 따라 나노입자 크기의 증가와 응집현상을 관찰할 수 있었다. 최적온도와 최적광량은 각각 40℃ 그리고 100 μmole·m-2s-1로 판명되었다. 합성된 금속 나노입자들의 투과전자현미경 관찰결과 은과 금 나노입자의 평균 직경은 각각 13 nm 그리고 39 nm 이었다. 은 나노입자의 경우 대부분 구형이었으나, 금 나노입자의 경우 대부분 불규칙한 형태로 관찰되었다. 격자구조d-spacing 값의 전자 회절분석, 제한시야 전자회절 패턴분석 그리고 엑스선 회절분석을 통하여 (111), (200), (220), (311)의 회절 패턴을 확인하여 전형적인 금과 은 나노입자의 결정구조 존재를 확인하였다. 합성된 은 나노입자는 대표적인 그람 음성 및 양성 균주 4종에 대하여 항균력을 보였으며, 그 중에서 Bacillus subtilis가 가장 민감하였다. 그러나 금 나노입자의 경우 항균력을 보이지 않았으며, 이는 나노입자의 크기와 표면개질로 인한 것으로 사료된다. 본 연구결과를 통하여 대표적인 바이오매스인 목재 추출물이 금과 은 나노입자 합성에 효과적으로 활용될 수 있으며, 향후 두 금속 나노입자의 대량생산도 가능할 것으로 사료된다.

In this study, phytosynthesis, characterization, and antibacterial efficacy of silver and gold nanoparticles (NPs) produced by hot water extract of mixed woodchip powder has been investigated. Composition and elemental analysis of the woodchip powder showed the presence of various sugars originated from cellulosic and hemicellulosic biomass and there was no significant amount of toxic heavy metals. The woodchip extract was successfully used as a reducing and stabilizing agent for the biosynthesis of the crystalline metal NPs. Effect of different factors affecting the metal NPs formation were evaluated based on the measurements of surface plasmon resonance (SPR). In the case of AgNPs, gradual increase in absorbance of SPR band was observed as the increase of AgNO3 and woodchip extract concentration. The formation of AgNPs was significantly increased at alkaline pH (>11) and a blue shift in the SPR band was observed as the increase of the reaction pH. The optimum temperature and strength of visible light irradiance were 30℃ and 100 μmole/m2/s, respectively. In the case of phytosynthesis of AuNPs, the formation was facilitated in between weak acidic and neutral pH 5-7 and a blue shift in the SPR band was also observed as the increase of reaction pH. Evident SPR band (546 nm) was not observed when the concentration of HAuCl4 was increased more than 0.5 mM and neglectable change in SPR spectra was observed in response to the concentration of the woodchip extract. The optimum temperature was determined as 40℃ and the blue shift phenomenon was also observed as the temperature increased. The optimum light energy input was 100 μmole/m2/s in our experimental condition. High resolution transmission electron microscopy of the metal NPs showed formation of well dispersed and mostly spherical Ag- and AuNPs with the average diameters of 16 and 6nm, respectively. The crystalline nature of the both NPs was analyzed by calculating d-spacing between lattice structure of the NPs and bright circular ring pattern from selected area electron diffraction analysis which can be indexed (111), (200), (220), and (311) facets of silver. In addition, obvious diffraction pattern was confirmed by X-ray diffraction analysis. Elemental composition analysis of the NPs using an energy dispersive X-ray spectroscopy indicates involvement carbonic biomolecules from the woodchip extract in the formation of the NPs. The phytosynthesized AgNPs showed clear antibacterial activity against four bacterial strains. However, the AuNPs lost its bactericidal activity which might resulted from surface modification of the NPs. These results suggest that the phytosynthetic method of the metal NPs using woodchip extract is highly effective. In addition, the AgNPs can be used as an antibacterial agent and the AuNPs could be useful for clinical and biomedical applications.
