Background: Identifying stress vulnerability after antidepressant discontinuation can be useful in preventing depression recurrence. Previous study demonstrated that a part of the innate immune system within the central nervous system (CNS), microglia was involved in the stress vulnerability. Thus, I hypothesized that persistent glucocorticoid (GCs) exposure by chronic stress might be involved in microglia functional properties, leading to stress vulnerability. Methods: First, the mice were exposed to chronic restraint stress (CRS) for 21 consecutive days (2h/day) with or without co-treatment of imipramine. Subsequently, these groups were exposed to a restraint stress (RS) as additional stress after imipramine washout. To assess depression-like behaviors, sucrose preference test, light dark test (LD), elevated plus maze (EPM), tail suspension test (TST), and forced swim test (FST) were performed after CRS. Main targets of stress and antidepressants were analyzed in the hippocampus after series of depression-like behavior analysis. In vitro study, BV2 murine microglial cell line and primary cultured microglia were used to examine the change of functional properties to a glucocorticoid stimulus. After glucocorticoid receptor agonist (dexamethasone, DEX) treatment in BV2 cell line and primary cultured microglia, the microglia was isolated for qPCR, western blot, and immunofluorescence to investigate possible mechanism on change of microglia function to glucocorticoid stimuli. Result: We found for the first time that mice exposed to CRS with imipramine co-treatment, which did not show depressive-like behaviors, were vulnerable to subsequent RS stimuli compared to the non-stressed mice after washout period. Imipramine did not rescue anti-inflammatory microglial phenotypes in the hippocampus of CRS mice although it restored depressive-like behaviors. We found that 72h of DEX treatment reduced fractalkine receptor (CX3CR1) and CD200 receptor (CD200R) in BV2 murine microglia and primary cultured microglia. In addition, the effect was abolished by RU486 (GC antagonist) co-treatment, suggesting that glucocorticoid receptor mediates the dexamethasone effect on CX3CR1 and CD200R reduction in microglia. We found that the DEX effect on anti-inflammatory microglia phenotype might be mediated by yy1-histone deacetylase 2 (HDAC2) signaling. Conclusion: Overall, our results suggest that chronic GCs exposure may reduce anti-inflammatory-like phenotype of microglia (CX3CR1 and CD200R) via their specific pathway (like yy1-HDAC2), which may be involved in stress vulnerability and depression.
