Background: Stress, by itself, is not only a factor causing neuropsychiatric disorders such as depression and anxiety, but it is also known to play an important role as a deterioration factor on neurodegenerative diseases such as Alzheimer’s disease. Despite microglia have attracted recently major attention in these neurological diseases, there are still few studies on the effect of stress hormones on the microglia and their association with diseases. Methods: To address this issue, 2-day-old Sprague Dawley (SD) rats were sacrificed for primary microglia culture. To investigate the effect of glucocorticoids (GCs) on microglia, dexamethasone (DEX), GCs receptor agonist, was treated in primary cultured microglia and the functional characters of microglia were analyzed using immunofluorescence (IF), western blot (WB), qPCR, and phagocytosis assay, and transcriptome analysis. Result: In this study, DEX inhibited the most of intrinsic functions of microglia such as cytokines secretion, phagocytosis function and proliferation function despite DEX induced ramified form of morphology. The decrease of microglial signature markers were sustained despite DEX washout. In transcriptome analysis, we conformed that DEX-treated microglia was close to senescence form. Conclusion: Our data suggest that stress hormone can lead to microglia dysfunction and it seems to be irreversible or persistent. Considering that microglia is a dynamic cell on maintaining homeostasis in CNS, DEX-induced microglia dysfunction might be involved in pathogenesis of neuropsychiatric disorders and neurodegenerative disorders. Additionally, we propose that targeting microglia senescence might be a new therapeutic strategy in neurological disorders.
