A subpopulation of mice exhibiting addiction-like behavior are selected out from cocaine self-administered animals, and the heightened level of D2R abundance is observed in cholinergic interneuron of these mice but not from the other cocaine-exposed mice. Higher D2R abundance in cholinergic interneurons is sufficient to induce higher motivation to cocaine, which suggest that animals are more vulnerable to addiction. Moreover, chronic overexpression of D2R in cholinergic interneurons is sufficient to mimic the cocaine-induced neural plasticity such as increased spine density, decreased membrane excitability, and input-specific synaptic plasticity that selectively occurs in the dopamine-D1-receptor-expressing medium spiny neurons (D1-MSNs). Finally, I find that cholinergic interneurons have differential connectivity onto D1- compared to D2-MSNs, which can putatively play a pivotal role in the cell-type specific alteration of MSN. These findings reveal an unknown mechanism of drug addiction, which may also implicate for loss of cognitive flexibility and compulsive behavior. Collectively, this study provides evidence that dopamine contributes to a synaptic plasticity of an amygdala inhibitory circuit underlying appropriate expression of less-salient fear memory. In addition, modulation of abundance of D2R that regulates the neuronal activity of accumbal cholinergic interneurons can mimic cocaine-induced neuroadaptations, which may result in rapider or easier lapse into severe stage of drug addiction.
