Hyaluronan에 대한 면역형광염색, CD44, PCNA, filaggrin에 대한 면역조직화학염색도 시행하였다. 이와 함께 배양된 각질형성세포에 IPL을 조사 후 24시간, 48시간에 세포와 supernatant를 얻어 hyaluronan에 대한 ELISA 및 CD44, HAS3 mRNA에 대한 real time RT-PCR을 시행하였다. 연구 결과 IPL과 고주파는 young mice와 aged mice 모두에서 표피 내 hyaluronan의 발현을 증가시켰으며 이는 ELISA, RT-PCR, 면역조직화학염색 모두 동일한 소견을 보였다. 단, IPL은 시술 1일 후에 가장 발현량이 많았던 반면, 고주파는 시술 3일 후에 가장 발현량이 많았다. IPL과 고주파는 hyaluronan synthase3과 수용체인 CD44 발현을 증가시켰으며 이는 ELISA와 면역조직화학염색 모두 동일한 결과를 보였다. IPL과 고주파는 aged mice에서 표피증식의 표지자인 PCNA 발현과 표피분화의 표지자인 filaggrin의 발현을 증가시켰으며, 이러한 발현 증가는 시술 7일까지 통계적으로 유의하게 유지되었다. 또한 배양된 각질형성세포에 IPL을 조사하였을 때 hyaluronan, hyaluronan synthase3, CD44 모두 발현이 증가하는 것을 확인하였다. 결론적으로 photorejuvenation 장비인 IPL과 고주파는 기존에 알려진 진피 내 콜라겐 및 탄력섬유의 합성 증가뿐 아니라, 표피 내 hyaluronan 발현을 증가시킴으로써 각질형성세포의 증식 및 분화를 촉진하여 노화된 피부의 표피기능 이상을 호전시킬 수 있음을 확인할 수 있었다.

Yet, the cellular and molecular mechanisms that cause epidermal dysfunction during skin aging are not well understood. Recently, the role of epidermal hyaluronan (HA) as an active regulator of dynamic cellular processes is getting attention and alterations in HA metabolism are thought to be important in age-related epidermal dysfunction. Intense pulsed light (IPL) and microneedle fractional radiofrequency (RF) has shown effects for improving cutaneous aging. However, little is known about the effects of IPL and fractional RF on the epidermal HA and epidermal function. We investigated the effect of IPL and fractional RF on the expression of epidermal HA and hyaluronan synthase (HAS) in young and aged mice epidermis and cultured keratinocytes. We performed IPL and fractional RF on the dorsal skin of thirty 8 week-old (young) hairless mice and fifteen 47 week-old (aged) C57BL/6J mice. Skin samples were collected on day 1, 3 and 7. HA content was measured by ELISA. Gene expressions of hyaluronan binding protein 4 (HABP4), CD 44, and HAS3 were measured using real time RT-PCR. Immunohistochemistry for detection of HA, CD44, PCNA and filaggrin were performed. We also investigated expression levels of HA and gene expressions of HAS3 and CD44 after IPL irradiation in cultured human keratinocytes. HA content and the mRNA levels of HABP4, CD44, and HAS3 were upregulated in the epidermis of both young and aged mice after IPL and fractional RF treatment. The expression was increased from day 1 after treatment and increased expression persisted on day 7. In addition, IPL increased HA content and mRNA levels of HAS3 and CD44 in cultured human keratinocytes. IPL and fractional RF treatment significantly increased PCNA and filaggrin expression only in the aged mice skin. IPL and microneedle fractional RF increased epidermal HA and CD44 expression in both young and aged mice and reversed age-related epidermal dysfunction especially in aged mice, suggesting a new mechanism involved in the skin rejuvenation effect of IPL and microneedle fractional RF.
