Laurus nobilis which belongs to the family Lauraceae is widely cultivated in Europe and Middle East. In Iranian folk medicine, L.nobilis leaves has been used for rheumatic pains, treatment of muscular convelsion. Recent studies have shown that the leaves of L. nobilis exhibited a significant analagesic and anti-inflammatory effect in tail-flick and formalin tests. In the searching of anti-inflammatory compounds from natural products, methanolic extract of L. nobilis leaves was partitioned with n-hexane, CH2Cl2, EtOAc, n-BuOH and H2O. Further fractionation of CH2Cl2 and EtOAc -soluble fractions resulted in the isolation of two new sesquiterpene lactones, 3-peroxyreynosin (6) and laurenoperoxyolide (7), a new phenolic compound, laurenodiol A (22) together with nineteen sesquiterpene lactones, santamarine (1), magnolialide (2), armefolin (3), 3-peroxyarmefolin (4), reynosin (5), costunolide (8) 11-exo-methylensantonin (9), tuberiferin (10) sivosinolide (11), altissin (12), maroniolid (13), 1-epi-tatridin B (14), a germacranolide with an 1,5-ether linkage (15), zaluzanin C (16), zaluzanin D (17), dehydrocostus lactone (18), eremanthine (19), 4α-hydroxy-guaia-10(14),11(13)-diene-12,6α-olide (20), spirafolide (21), three phenols, 1-(4-hydroxy-3-methoxyphenyl) propan-1,2-diol (23), homovanillic alcohol (24), syringaldehyde (25), two monoterpenes, 8-hydroxy-p-menth-6-en-2-one (26), 6-hydroxycarvotanacetone (27), one megastigmane, (-)-loliolide (28), three coumarins, 7-hydroxycoumarin (29), grevillon (30), isoimperatorin (31), five flavonoids, (+)-catechin (32), (-)-epicatechin (33), kaempferol (34), kaempferol-3-O-glucoside (35) and kaempferol-3-O-α-L-rhamnoside (36). Their structures were elucidated on the basis of spectroscopic methods such as 1D-NMR (1H, 13C), 2D-NMR (HSQC, HMBC, COSY, NOESY) and HRESI-TOF-MS. Compounds 1, 2, 5, 9, 13 and 17 potently inhibited NF-κB with IC50 values of < 15 μM. The results showed that the leaves of L. nobilis might be developed as anti-inflammatory product.
