Material and Methods 6-hydroxydopamine (6-OHDA) was injected into the medial forebrain bundle of rat. Two weeks later. PD rat model was verified by apomorphine-induced rotation test. Two weeks later, experimental animals were divided following two groups :(1) the group were injected with adeno-associated virus carrying halorhodopsin (mPV-eNpHR-YFP AAV) (n = 5), (2) the group were injected with adeno-associated virus carrying only the YFP gene (AAV2-mPV-YFP) (n = 5). The optic fiber was implanted into the ipsilateral endopeduncular nucleus after 2 weeks. A stepping test were performed in 3 sequential steps: during light-off state, during light stimulation, and again during light-off state. The light stimulation protocol was (width, light frequency) followed: (1) a width of 10 ms and a frequency of 5 Hz, (2) a width of 100 ms and a frequency of 5 Hz, and (3) continuous. And then, the PD rat was sacrificed. immunohistochemistry(IHC) study was done.
Results In the group using adenovirus carrying the halorhodopsin gene, akinesia was statically improved comparing control group comparing with control group. (p value =0.000) Also, in optic stimulation, the number of stepping were improved than that in non-stimulation. (p=0.000) In IHC, loss of tyrosine kinase in substantia nigra confirmed the PD rat model. And YFP and GAD67 were stained well in EP, it confirmed that the optogene in EP was expressed by the injected virus (adeno-associated virus carrying halorhodopsin, mPV-eNpHR-YFP AAV)
Conclusion In the model of Parkinson 's rat model, it was confirmed that the inhibition of EP using optic stimulation after halothdorp implantation can improve the motor ability for the first time. In the future, the possibility of the treatment of Parkinson' s disease by optical stimulation is suggested by this study
